Objective:To explore the effects of traffic related PM2.5on the cytokines in peripheral blood and Ca2+--CaN/IRAK-1-NFAT signal pathway regulate cytokines, to provide the reference basis of investigation mechanism of the immunotoxicity of PM2.5.Methods:This study includes two populations:traffic polices of Taiyuan as the objective group and the workers of Disease Control and Prevention Center (CDC) as the control group. The concentration of PM2.5in their own working atmosphere was monitored. Their peripheral blood was collected and separated lymphocytes. The intracellular calcium concentrations ([Ca2+]i)were measured by fluorescence spectrophotometry; the expression levels of tumor necrosis factor a (TNF-a), interleukin-1(IL-1), interleukin-2(IL-2), interleukin-4(IL-4) in serum were detected by ELISA. The fluorescence quantitative PCR (Q-PCR) was used to determine the mRNA levels of calcineurin (CaN), interleukin-1receptor-associated kinase (IRAK-1), Nuclear factor of activated T cells (NFAT), IL-2and TNF-a. Western Blotting was used to determine the protein levels of CaN, IRAK-1and NFAT.Results:The concentration of PM2.5in traffic polices’working atmosphere was higher than control group (0.132mg/m3vs0.100mg/m3)(P<0.05).[Ca2+]i of traffic police was higher than control group.The mRNA levels of IL-2in objective group was higher than control group (P<0.05). The mRNA levels of IL-2in male traffic police was higher than the male worker of CDC (P<0.05).The levels of TNF-a and IL-1in serum of objective group were higher than control group (P<0.05), however, the levels of IL-2in serum of objective group was lower than control group (P<0.05). Compared to control group, the mRNA levels of CaN and IRAK-1in traffic police appeared a raise trend and the mRNA levels of NFAT was higher (P<0.05). There were positive correlation between the level of TNF-a and [Ca2+]i, CaN mRNA, NFAT mRNA expression respectively. Meanwhile there were negative correlation between IL-2and Ca2+However, no siginficant correlation could be found between IL-1and Ca+, CaN mRNA, IRAK-1mRNA, NFAT mRNA.Conclusion:Traffic related PM2.5in Taiyuan could increase the expression of IL-1and TNF-a in peripheral blood and suppress the secretion of IL-2. The Ca2+-CaN/IRAK-1-NFAT signaling pathway of peripheral blood lymphocyte may involve in the regulation of TNF-a.
