Objectives: To investigate the correlation between TPA and the pathogenesis of preeclampsia by determining concentration of tissue polypeptide antigen in serum from pre-eclamptic and normotensive pregnant (NP)women, and watch whether the serum from pre-eclamptic women can induce the expression of vascular cell adhesion molecule-1 on human endothelial cells in vitro .Methods: The experiment was divided into the following two parts.First, ELISA was used to quantitate the concentration of TPA, sVCAM-1, sE-selectin in serum The relation of them were analyzed by rank correlation.Second,Human umbilical veil endothelial cells were incubated with medium containing 20% calf serum,which were seeded in a 24-multiwell flat-bottomed microtiter plate.They were stimulated by serum from pre-eclamptic and normotensive pregnant women, then the expression of VCAM-1 was evaluated by indirect immunofluorescence microscopy.Results : (1)The level of TPA, sVCAM-L sE-selectin in materal peripheral blood of pre-eclamptic patients was significantly higher than NP women.There were significant difference between the two groups(P<0.05) But there was no difference of them between mild and severe pre-eclamptic patients (P>0.05).(2)There were positive correlation among TPA,sVCAM-1 and sE-selectin level (The relative coefficient respectively was 0.419, 0.659).(3)Compared with NP women, the serum from severe pre-eclamptic women obviously stimulated the expression of VCAM-1 on HUVEC (P<0.05) But there was no difference between the serum from mild pre-eclamptic and NP women in increasing the expression of VCAM-1. (4)The concentration of TPA, sVCAM-1, sE-selectin was positively related to the serious degree of preeclampsia and negatively related to the fetal weight,especially in the severe pre-eclamptic women.Conclusion: (1) The level of TPA in materal peripheral blood was much higher in pre-eclamptic patients and the correlation between the TPA and adhesion molecules showed that STBM may be a toxic factor, which damaged the endothelial cell. The serum from severe pre-eclamptic patients obviously increased the expression of VCAM-1 on HUVEC.It could suggest that the serum of high concentration of TPA could arouse the damage of HUVEC. (2)The concentration of TPA, sVCAM-1 , sE-selectin was positively related to the serious degree of preeclampsia, and negatively related to the fetal weight TPA may be regarded as the index of progress and monitor of preeclampsia .
