Objective: To research the influence of diazoxide to apoptotic geneexpression of rat myocardial cell induced by isoprenaline .Methods: 32 male rats (Sprague-Dawley) were allocated to four groupsrandomly, 1. Normal group(Nor, control group) ; 2. Isoprenaline group ( Iso, toinduce cell apoptosis); 3. Diazoxide group (DZ, mitoKATP channel opener); 4.Glibenclamide group (GL, mitoKATP channel inhibitor).The relative quantity ofBcl-2 and caspase-3 expression of each group were measured by RT-PCR( reversetranscription polymerase chain reaction ) technique. Myocardium cell apoptosis ofeach group was tested by TUNEL(terminal-deoxynucleotidyl transferase mediatednick end labeling) technique and the apoptotic index (AI) was calculated .Result: The relative quantity of Bcl-2 expression of DZ group is higher thanIso group and GL group (p<0.05).But the relative quantity of Caspase-3 expressionof DZ group is lower than Iso group and GL group (p<0.05) . There is no significantdifference on the relative quantity of Bcl-2 and Caspase-3 expression between Isogroup and GL group . The p value is 0.134 and 0.297 respectively . The AI of DZgroup is lower the Iso group and GL group (p<0.01). The AI between Iso group andGL group has no significant difference (p=0.156) .Conclusion: The diazoxide can protect myocardium cell from apoptosisinduced by isoprenaline. The mechanism may be that diazoxide can up-regulationthe Bcl-2 expression and down-regulation the caspase-3 expression.
