Objective: To investigate the effect of mitochondrion on apoptosis medicated by carbondisulfide in testicular germ cell in male rats.Methods: Twenty four SD male rats were randomly divided into four CS2treated groups.Rats were exposed to multiple levels of CS2concentration (0,50,250,1250mg/m3) byinhalation for10weeks,5d/w,2h/d. After treatment, rats were sacrificed by decapitation.Cytoplasm protein in testis was extracted for detecting the levels of AIF, Cyto C, Bcl-2, Bax,Procaspase-9/3by western blot. Mitochondrial protein was extracted to detect the level ofMPTP and mitochondrial respiratory chain enzyme complex I-IV by enzyme-linked immunesorbent assay. Protein in testis was extracted for testing the activities of Caspase-9/3. RNAwas extracted, and qPCR detection was performed to explore the gene expression levels ofAIF, Cyto C, Bcl-2, Bax, Smac, Procaspase-9/3in testicular germ cell.Results: Compared with the controls, levels of Cyto C in cytoplasm protein in testiculargerm cell increased when rats were exposed to different doses of CS2(P <0.05). The ratiobetween Bax and Bcl-2, and the activities of Caspase-9/3increased significantly, while thelevels of Procaspase-9and Procaspase-3decreased in moderate concentration CS2treatmentgroup (P <0.05); high dose of CS2exposure increased the levels of Bax and AIF, anddecreased Bcl-2level in testicular germ cell, these changes were all significant (P <0.05).The level of MPTP showed significant decreasing in each group compared with thecontrol group (P <0.05). Meanwhile, levels of respiratory chain enzyme complex I-IV intesticular germ cell were increased when rats were exposed to different doses of CS2(P<0.05). The mRNA levels of Caspase-9and Bax in testicular germ cell, as well as the ratiobetween Bax and Bcl-2, were significantly up-regulated after rats were treated with CS2(P <0.05). When rats were exposed to a high dose of CS2, mRNA levels of Cyto C, Caspase-3and AIF showed significant increasing, while that of Bcl-2was significantly decreased (P <0.05).Conclusion: CS2exposure could affect the expression of protein and gene which is related tomitochondrial mediated apoptosis in testicular germ cell. Meanwhile, it could impact on theactivities of some proteins, as well as the normal oxidative phosphorylation inmitochondrion, obstruct the progress of ATP synthesis, and consequently, result in apoptosisin testicular germ cell in male rats.
