Objective:12-lipoxygenase(12-LOX)was found to have a variety ofthe biological behavior through multiple signal pathway,but little wasknown concerning its mechanism. We studied the effect of the selectiveinterruption on the angiogenesis and apoptosis inducement in gastriccancer cells by treating with Baicalein.Methods:The variation of VEGF vascular endothelial growth factorexpression was detected through verse transcription-polymerasechain(RT-PCR) and immuocytochemistry staining.The synergistic effectsof Baicalein on the proliferation and apotosis of gastric cancer cell werestudied by using MTT assays,acridine orange and ethidium brom ides (AO/EB)stains and in situ cell apotosis detection.Results:Baicalein assumes the concentration dependent manner tosuppress SGC-7901 cell VEGF mRNA and the protein expression;Differentdensity Baicalein has the inhibitory action to the tumor cellproliferation in a time and concentration dependent;Baicalein can alsoinduce apotosis on the gastric cancer cell which the density and theresponse time are related;Conclusion: 1.Blocks the 12-LOX pathway to be possible to suppressVEGF production function through the decline gastric cancer cell VEGFexpression.2.The fuction of proliferation suppression and apoptosisinducement assumes the certain time and the dosage.3. Cell apoptosisinduced by Baicalein may be related to the decline VEGF to concern.
