Objective:To investigate the Effect of 12-lipoxygenase(12-LOX) on pro- liferation and apoptosis of gastric cancer cell lines AGS.Methods: AGS cells cultured in vitro were as a model. 12-LOX mRNA was analyzed by RT-PCR;Baicalein, the specific 12-LOX inhibitor with con- centrations from 20 to 80μmol/L and 12-HETE, the 12-LOX product with concentrations from 100 to 400nmol/L were treated in AGS cells from 24 to 72h.Morphological changes on cellular and subcellullar structures were observed and photographed with inverted microscope and electron micro- scopy;The proliferation was detected by trypan blue exclusion method and MTT(thiazolyl blue tetrazolium bromide)methods;The apoptosis was mea- sured by AO-EB fluorescence staining,TUNEL staining and DNA-fragment assay,respectively.Results:12-LOX mRNA was expressed in Human AGS cell lines;Morphological changes such as chromatin condensatio, apoptitic bodies were observed. Concentrations from 100 to 400 nmol/L,12-HETE can promote proliferation and inhibite apoptosis from 24 to 72h in a time and concentration de- pendent way approximately(P<0.05vs control). Within 72h the maximal survival rate was 111.52%,while the minimal one was 101.39%.Concen- trations from 20 to 80μmol/L,baicalein inhibited proliferation and induced apoptosis from 24 to 72h in a time and concentration-dependent way approximately(P<0.05 vs control).Within 72h the maximal survival rate was 95.83%,while the minimal one was 38.90%.Conclusion:12-LOX mRNA was expressed in AGS cell lines.12-HETE can promote proliferation and induce apoptosis of human gastric cancer cells...
