[Abstract]Objection To evaluate the feasibility of regeneratingor repairing damaged skin utilizing mesenchymol stem cells by tissueengineering techniques.Materials and Methods Mesenchymol stem cellswere isolated from New Zealand rabit of 1 month,cultured with DMEMcomplete medium in vitro. MSCs of 3~5 passages were seeded on porousfoam PLGA scaffolds after marked by DAPI,and then observed by scanningelectron microscope and fluorescence microscope. The skin of 2×2cm2were cut from the back of rabbit, and then the compound of MSCs-PLGAand the porous foam PLGA scaffolds alone were tansplanted on itrespectively. After operation, wound healing was observed, HE stain,Van-Gieson stain and immunocytochemistry of cytokeratin AE1/AE3 wereperformed to detect the transplants after 5 weeks. Results After 3 daysof cultivation, most of the MSCs adhered the wall. At the 10th day ofcultivation, cells grew confluently and then passaged. Both scanningelectron microscope and fluorescence microscope showed that cells grewwell on porous foam PLGA scaffolds, and the number of cells increasedgradually. Animal experiment showed that with the degradation ofbiodegradable polymer scaffolds,the skin was gradually replaced by thecompound of MSCs-PLGA, HE stain showed the structure of the new skinwere as the same as the normal skin,the cells of blue Fluorescentnucleus could been found in fluorescence microscope andimmunocytochemistry showed cytokeratin expressed in cutinized layerand hair follicle.But the skin repaired by porous foam PLGA scaffoldsshowed little hair follicle and thickening fibrosis scar. ConlusionsMesenchymal stem cells can be successfully isolated and cultured byadhering to the culture plastic. No obvious cytotoxicity was observedin PLGA polymer and it is a good carrier for MSCs. The damaged skincan be repaired utilizing the compound of MSCs and PLGA polymer.
