Objective: 1. To study the synthesis of N-[2-(cholester-yloxycarbonylamino) ethyl]carbamoylmethylated mannan ( chol-AECM-mannan) . 2. Experimental study of mannan-modified targeted nanoliposome for cancer biotherapy.Methods: 1. Synthesize a compound with polysaccharides partially modified by cholesterol(namely chol-AECM-mannan) through carboxymethylating, carbamoylating and condensation reaction. The main material is mannan. 2. 3 β [N-(N', N' -dimethylaminoethane) -carbamoyl] cholesterol (DC-chol) has been synthesized by cholesteryl chloroformate and N,N- dimethylethylendiamine, and then used to prepare sonicated mannan-modified targeted nanoliposome withdioleoylphosphatidylethanolamine(DOPE) and chol-AECM-mannan. Measure the size of nanoliposome. Several nanoliposomes and purified EGFR are performed in those mice model of lung cancer made by implanted tumor pieces. And then observe those commonly status and tumors' growth; Measure the antibody titre in the blood serum of those mice by ELISA and Western.Results: 1. Confirm the product by IR spectrum and ~1H nuclearmagnetic resonance (!HNMR). And the proton integral ratio of mannan and cholesterol is 100 : 5, that is to say, the numbers of the substituted cholesterol per 100 mannose units are 1.1. The production rate is 24%. 2. The size of liposome nanoparticle is 132nm by zetasizer. Antibody is produced in the blood serum of the experimental group and its titre is far higher than the others ' by ELISA and Western. There is a statistic signification in term of volume of the mouse tumor. Tumor volume of the experiment group is far less than collator' (purified EGFR and null liposome) (P<0.05) .Conclusion: l.The method is a effective pathway in term of synthesizing chol-AECM-mannan. 2. It is effective on lung cancer in term of mannan-modified targeted nanoliposome.
