Objective To study the therapeutic results and mechanism of Epimedium on preventing ovariectomy-induced osteoporosis and expression of TNF - α, TGF -β1 in microenvironment of vertebra in rats.Methods 45 female SD rats at 3 months were randomly divided into control group (group 1), tendency group (group 2) and experiment group (group 3). Groups 2 and 3 all been amputated ovary, group 3 were given Epimedium extract. Group 1 were given to use skill handling. Termination of 5 rats in all groups took place respectively after operation 4 weeks, 8 weeks and 12 weeks. The 5th lumbar vertebra was taken and paraffin section was made. The samples were dyed with HE, Van Gieson and immunohistochemical stain of TNF - α and TGF-β1. Bone little beam structure were analysed with Image Analyst, and multi-nuclei / osteoclast like-cells were counted.Results Compare with control group, Tb.Th and Tr.Ar have obvious decreasing while Tb.Sp increasing in tendency group. Trabecular bone in Epimedium group measure is similar to compare group. The number of multi-nuclei / osteoclast like-cells in tendency group is obviously more thangroup 1 and group 3. Expression of TNF-a showed that positive cells of monocytes in group 2 and weak positive or negative cells in group 1 and group 3. Osteoblasts and matrix of group 1 and 3 showed positive reaction of TGF-Pi but negative in group 2.Conclusion In this study, we successfully made a model of postmenopausal osteoporosis by ovariectomy. Epimedium can inhibit bone loss early and prevent efficiently the ovariectomy-induced osteoporosis. Epimedium obviously inhibit osteoclasteogenesis and bone resorption by down-regulate the expression of TNF-a, and induce the expression of TGF-Pi in microenvironment. We suppose that flavonoids, which the main component of Epimedium, can result in this effect because it4ias resemble chemistry structure as estrogen.
