Epimedium, also known as Xianlingpi in china is the dried leaves of Epimedium brevicornu Maxim.,Epimedium sagittatum(Sieb.et Zucc.)Maxim.,Epimedium pubescens Maxim.,Epimedium koreanum Nakai(family berberidaceae), which is provinces of Shanxi,Liaoning,Shanxi,Hubei,Sichuan,etc. It is a traditional Chinese tonic medicine, it is pungent and sweet in flavor,warm in property,acting on the liver and kidney channels.The effect of tonifying the kindey,strengthening yang,dispelling wind and eliminating dampness.The unprepared epimedium effect emphasis on dispelling wind and eliminating dampness, epimedium after stir-baked with sheep fat can improve the effect of tonifying the kindey,strengthening yang. This item is supported by the research "The study on normative using19Chinese medicine"(No.20110700717). We will undertake the research on different between unprepared epimedium and processed epimedium, providing theoretical basses on "difference between using unprepared epimedium and processed epimedium".The thesis is divided into four chapters.Systematic and overall researches on traditional processing way of epimedium would be executed by consulting a lot of archaic medicine works and modern processing books。After the organize of the literatures,we found that the sheep fat-baking way was most widely used for epimedium processing way not only in ancient times but also in modern. The above conclusion provide theoretical supports for processing epimedium by sheep fat-baking. Conducted a questionnaire survey on the clinical use of unprepared epimedium and sheep fat-baking epimedium, The results showed that romiscuous using between epimedium and sheep fat-baking epimedium.Select the optimal processing technics of epimedium,The main influential factors of processing effect included the processing temperature and time, the content of Icariin and Baohuoside I was used as the assessment index, the optimal processing conditions were defined by Central Composite Design-response Surface Methodology. The optimum conditions were added the epimedium after melting sheep fat and control the temperature of160℃,stir for7minutes.20kg sheep fat was used for per100kg epimedium. This processing method was proven that it’s stable and feasible by checking experiment.Research the changes between unprepared epimedium and sheep fat-baking epimedium by thin layer chromatograph,high Performance Liquid Chromatography and HPLC fingerprint. The results show that relative to unprepared epimedium, there was no new peaks appear and original peaks did not disappear in the fingerprint,8peaks declined and8peaks increased obviously,the content of Epimendin Al increased18.2%and Icariin increased13.7%, Epimendin B decreased by47.3%and Baohuoside I decreased by26.8%,and the content of Epimendin C not changed very much. In summary, there was superior difference with chemical constituents between unprepared epimedium and sheep fat-baking epimedium.To study the pharmacokinetics of unprepared epimedium and sheep fat-baking epimedium in rats, the rats were divided into two groups randomly and received an intragastric administration of unprepared epimedium and sheep fat-baking epimedium, The plasma sample were collected as scheduled and icariin were analyzed by HPLC-MS.The Pharmacokinetic parameters of icariin were calculated and evaluated by DAS2.0.The results show that in epimedium group, the Cmax and AUC(O-t) of were2.285±0.75mg·L-1and14.935±5.11mg/L·h, while, the Cmax of processed epimedium group was2.8±0.82mg·L-1and the AUC(0-t) was25.301±8.12mg/L·h. The oral bioavailability of icariin in epimedium has been enhanced in rats after processing. In summary,there are some changes in chemical composition and pharmacokinetics between unprepared epimedium and sheep fat-baking epimedium.And these changes may cause the change in the efficacy of unprepared epimedium and sheep fat-baking epimedium.
