Objective: To investigate the impact of single nucleotide polymorphism 43(UCSNP 43) of calpain-10 gene(CAPN-10) on NIDDM1 locus, chromosome 2q37.3 on polycystic ovary syndrome(PCOS) with or without Insulin resistance(IR). Methods:Case-control study. Ninety-six PCOS and 96 control subjects were included in the study. Endocrinal hormone,oral glucose tolerance test(OGTT) and insulin were measured with radioimmunoassay, PCR-PFLP method was used to determine the distribution of allele and g enotype frequencies of S NP-43 polymorphism. Results: (1) The genetype frequencies of GG, GA, AA of SNP-43 in the control subjects(non-PCOS) were 0.760,0.230and 0.010 respectively; and the G allele frequency was 0.875 (2) the G allele and GG genetype frequencies in group of PCOS and PCOS-IR were higer than that in group of control subjects(non-PCOS) and PCOS-NIR, but the differerces were not significant;(3)the results of DHT,DHEA-S,insuliln(0,30,60,120,180mm) were statistically significant higher in GG than in GA+AA in all samples;and the results of DHT, insuliln(30,60,120min) were statistically significant higher in GG than in GA+AA in 96 control subjects .(4) the frequency of father-Baldness was significant higher in PCOS and PCOS-IR respectivelythan control and PCOS-NIR ; the frequency of acne was significant higher in PCOS than control; DHEA,assessment of body hair and BMI were significantly inseased in PCOS-IR as compared with PCOS-NIR.(5) hyperandrogenism and hyperinsulinism were the risk factors of PCOS;hirsutism,obese and father-baldness were the risk factors of PCOS-IR.Conclusion: G allele of SNP 43 of calpain-10 gene may contribute to hyperandrogenism and insulin resistance.
