Objective: Colorectal cancer is the most common malignant cancer. The present studies indicated that actived several oncogenes and inactived tumor suppressor genes are involved in colorectal cancer. The mutation of mismatch repair genes is also reported to be related to this process.We selected four genes which have significant different expression in cancer tissue compared to normal tissue from differentially expressed 86 genes by methods of suppression subtractive hybridization combined with cDNA microarray. They are gene of heat shock lOkDa protein 1 (HSPE1 HSP10) , SRY-box9 (SOX9), polymeric immunoglobulin receptor ( PIGR) and eukaryotic translation elongation factor 1 delta , EEF1 —delta.HSPE1 acts along with heat shock 60kDa protein 1 (HSPD1), help HSPD1to regulate functions of proteins by folding or extending proteins or assembling comlpex, make it possible to regulate proteins.The protein encoded by SOX9 gene recognizes the sequence CCTTGAG along with other members of the HMG-box class DNA-binding proteins, it acts as transcription factor. It acts during chondrocyte differentiation and, with steroidogenic factor 1, regulates transcription of the anti-Muellerian hormone (AMH) gene. Deficiencies lead to the skeletal malformation syndrome campomelic dysplasia,frequently with sex reversal.PIGR The poly-Ig receptor is expressed on several glandular epithelia including those of liver and breast. It mediates transcellular transport of polymeric immunoglobulin molecules. It is a member of the immunoglobulin superfamily. The receptor has 5 units with homology to the variable (V) units of immunoglobulins and a transmembrane region, which also has some homology to certain immunoglobulin variable regions. EEF1 — delta gene encodes a subunit of the elongation factor-1 complex, which is responsible for the enzymatic delivery of aminoacyl tRNAs to the ribosome.As has been mentioned, the four genes help to transcription and protein function and immunity actions. We analyse their expression in human rectum adenocarcinoma and normal rectum tissue. In order to discuss the mechanism of Human Rectum Adenocarcinomas.Method : Monitor the expression of HSPEK SOX9 and PIGR in patients who have human rectum adenocarcinoma through RT—PCR and Real-time PCR. Detect the expression of EEF1—delta in patients who have human rectum adenocarcinoma through Real-time PCR and line blot. Results:1. It has been showed that expression of the four genes in adenocarcinoma tissue has prominent difference compared with normal rectal tissue by the technique of Real—time PCR, HPSE1 and EEFl-delta show a higher exprssion in adenocacinoma;2. It has been showed that HSPEl and PIGR has prominent difference compared with normal by the technique of PT—PCR, while resultof S0X9 shows that they have nodifference;3. It has been showed that PIGR has no difference compared with normal by the technique of line blot.Conclusion: It has been showed thatthere are higher expression about HPSE1 and EEFl-delta in the primary adenocarcinoma tissue than in the normal rectum tissue. And at the same time, there are lower expression about S0X9 and PIGR in the primary adenocarcinoma tissue than in the normal rectum tissue.
