Background and purpose: Tetramine is a severe toxicant for killing mouse. Accidental and homicidal poisoning are common in both clinical and forensic aspects. But the intoxicated mechanism of tetramine is not known so far. GABA is the most primary inhibitive neurotransmitter in CNS. GABA_AR is one type of receptor for GABA, and the subunit α1 has significant relation to the function of GABAaR. Expression of GABA and GABAaR occur in response to a few of deseases and injury such as epilepsia, brain injury and intoxication. However the role of the GABA and GABAaR are not fully understood yet in rat poisoned by tetramine. To approach the intoxicated mechanisms we investigated the expression of GABA and GABA_AR-α1 in rat poisoned by tetramine by immunohistochemistry.Methods: sixty healthy Spragwe-Dawley rats were randomly divided into four groups including the control, the sham poisoned, the acute poisoned and the chronic poisoned. And the latter was once more subdivided into nine subgroups according to the sacrificed time of animals at 30min, 3h, 6h, 12h, 1d, 3d, 5d, 7d, 10d after poisoned. Each rat of acute poisoned group was given 1.0mg/kg tetramine by lavage and the rat of chronic poisoned group was given 0.1mg/kg tetramine by lavage. The expression of GABA and GABA_AR-α1were studied by immunohistochemistry and the results were assessed by image analysis system.Results: ?Moderate and ubiquitous but definite expression of GAB A and GABAaR-oiI was seen in the brain of control and sham poisoned groups. (2) The expression of GAB A and GABAAR-al decreased in the brain of acute poisoned group. ?The expression of GABA decreased at first in the brain of chronic poisoned group, and was lowest level in 6 hours after poisoned, then it enhanced and reached at peak in 5-7 days after poisoned, it was remain above the control till 10 days. @The expression of GABAaR-
