Objective: Sleep deprivation meant a state that the necessary sleepness was lost because of physiological, pathological or environmental reasons. With the crazy pace of modern society, as well as the requirement in some special departments, sleep deprivation became more and more common and unhealthy to human beings. By now, the drugs used to anti sleep deprivation usually act on center nerve system and would bring much badness when used a great deal or for a long time,which could not be used without permission from government. So, based on the study of the function of sleepness, the badness of sleep deprivation, the superiorities and shortcomings of the drugs used to treat sleep deprivation and the corresponding theory in Traditional Chinese Medicine, we developed a new drug made of ginsengoside and tea extraction: Xingshen Capsule. The objections of this article were as follows: to study the actions of Xingshen Capsule(No.l,2) against center and body fatigue; to reprography the model of sleep-deprived rats and investigate the protective effects of Xingshen Capsuleon on them; to explore the effecting mechanism of the new drug, and to define the optimal dosage of two main ingredients in this drug.Methods: 1. Mice, divided randomly into eight groups(control, KFY, XSNo.l low, middle, high, XSNo.2 low, middle, high ), were administered with different drugs once a day for six days .Then, their weight, motor activities, roating bar tolerance and the hypnogenetic effects of pentobarbital sodium on these mice wereobserved on the 1st and 6th day in order to see if XS could excite the mice and anti centre and body fatigue, and if the normal growth of the mice would be held back while taking XS at different dosages. 2."Modified multiple flower pots method" was used to set up 96h-sleep-deprived rat models. 3. Rats were divided randomly into six groups(normal,negative control, KFY, XSNo.2 low, middle, high ), and the animals were deprived of rapid eye movement (REM) sleep for 96 hours by the "modified multiple flower pots" technique except the normal group while administered with different drugs. Then, the refreshing effect of XS were studied by the general appearances, weights, activities in the "open field test" at 0, 24, 48, 72, 96 hours after sleep-deprivation, and the levels of serum Lactic Acid (LA), SOD, Cortisol (COR) , Testosterone (T) of these rats. COR and T were tested by radio immunoassay(RIA) while LA and SOD were tested by UV-spectrum. 3. Uniform-design method combined with pharmacodynamics was used to screening the optimal dose of ingredients in XS. Hypnogenetic effects of sodium pentobarbital and roating bar tolerance of the mice were observed in this experiment.Results: 1.Compared with the negative control, XS No.l and XS No.2 could notably increase the mice's motor activities, shorten the sleeping time caused by sodium pentobarbital while prolong its latency on both 1st and 6th day at middle and high dosages. The better effect could be seen at higher dosage and longer time. There was no significant difference between the middle and high dosage groups, but no effect was seen at low dosage. 2.Both XS No.l and XS No.2 could notably prolong the persisting time on the roating bar on both 1st and 6th day while No.2 seemed to be better. 3.Both of No.l and No.2 didn't interfere the normal growth of the mice in 7 days. 4.96h-sleep-deprived rat models were set up successfully by the "modified multiple flower pots" technique. 5.Compared with the sleep-deprivation group, XS could improve the energy metabolizing of sleep-deprived rats, hold back theirweight-decline, improve their cognize capability of space, and stabilize their movement in the open field. Besides, this drug contributed to meliorate the state of low-T and high-LA, SOD and COR caused by sleep-deprivation. 6.Accroding to the results of uniform-design, the most effective dosage of Ginsenoside used in XS was 2. 86mg/20g and for the extract from green tea ,14.19mg/20g. They could notably prolong the persisting time on the roating bar and shorten the sleep time caused by sodium pentobarbital.Conclusions: l.Xingshen Capsule might have optimistic effects on refreshing one's nerve, and the effect depends on the dosage and the time administered with the drug. 2.Xingshen Capsule might enhance physical strength and improve the capacity of body to energe-consumed actions. 3.Xingshen Capsule would not interfere the normal growth of mice. 4.96h-sleep-deprived rat models might be set up by the "modified multiple flower pots" technique. 5.XS might be a new drug made of Traditional Chinese herbs which do have definite protective effects in rats with sleep-deprivation. 6.The comprehensive effects on the modulating of nerve-endocrine- immunity network, HPA and HPG contribute to explain the mechanism of this drug. 7.XS is a new type of compound which used the extractions from Traditional Chinese herbs as its materials, and the dose of ingredients in it could be optimized based on their experiential dosages. 8.1t would be possible to use uniform-design combined with pharmacodynamics method in defining the optimal dosage of ingredients in XS.
