Objective: To study the realtionship between polymorphism Pro12Ala of PPAR-ygene and type 2 diabetes mellitus in xinjiang Uygur Kazak and Han population. Methods: in same population PCR-RFLP was used to determine the distribution of genotype and allele frequencies of Pro 12 Ala polymorphism in NGT (normal glucose tolerance) and T2DM(type 2 diabetes mellitus), the studied population were grouped according to the subgenotype, then the difference between each index was compared. Result: 1) in three population the frequencies of the Pro12Ala polymorphism in T2DM and NGT have no significantly difference. 2) the frequencied of "A" allele of the Pro12Ala was different in three population. The A allele in Uygurs and Kazaks was 0.1 land 0.09.similar to European,but was higher than Japanese and Han of shanghai. A allele in the Han of our study is similar to Japanese but is different to European. 3) in NGT of three population each indexes in the subgroup was no difference, but in T2DM, Ugyur population the A carrier has heavier waist and lower LDL-c than the P carrier. Compared with P carrier in Kazaks the waist and HDL were higher in A carriers. In Han population, the A carrier has more WHR and BMI. Conclusion: 1) in three population, the allele frequence and genotype frequencies did not differ significantly between T2DM and NGT, but the significant difference of Pro12Ala amont various races was observed. 2) PA+AA subgroup was associated with the abdominal in type 2 diabetes of Ugyur and Kazaks.and may impact their blood lipid. In Han population.PA+AA in type 2 diabetes may have related to fat and insulin resistance.
