Objective To investigate the effect of simvastatin on myocardial fibrosis and connective tissue growth factor(CTGF) in renovascular hypertensive rats.Methods The following groups of 24 Wistar rats were studied: Sham group rats, RH rats and RH rats treated with simvastatin (5mg.Kg-1.d-1.Systolic blood pressure (SBP) was measured.,collagen concentration (Coll) in terms of hydroxyproline concentration using Bergman method was also detected . VanGieson and immunohistochemistry staining ( SP method) combined with computed morphometty were used to evaluate the total collagen volume fraction (t-CVF), CTGF expression in left ventricular tissue.Results Compared with sham group, The SBP, t-CVF, Coll and CTGF expression increased markedly in RH group (P<0.01). Compared with the RH group, The SBP, t-CVF, Coll and CTGF expression significantly decreased in RH rats treated with simvastatin (P<0.05).Conclusion Simvastatin could inhibit the development of myocardial fibrosis in renovascular hypertensive rats ,Which was probably related to the decrease expression of cardiac CTGF.
