OBJECTIVE: The effects of sodium acetate, sodium butyrate, sodium caproate, sodium caprylate on the proliferation and apoptosis of MDA-MB-231 breast cancer cell line were investigated comparatively and the associated mechanisms were also discussed.METHODS: The effects and their mechanisms of sodium acetate, sodium butyrate, sodium caproate, sodium caprylate on anticancer in MDA-MB-231 breast cancer cell line were analyzed by cell culture, MTT-assay, flow cytometric analysis, invert microscope and transmission morphological examination, gel electrophoresis and immunocyto- chemical staining.RESULTS:①Sodium acetate (2.5×10-1mol/L~5×10-1mol/L), sodium butyrate (5×10-3mol/L ~1×10-1mol /L), sodium caproate(5×10-2mol/L~1×10-1mol/L) and sodium caprylate (5×10-3mol/L ~1×10-1mol/L) inhibit the growth of MDA-MB-231 breast cancer cell line in the time-dependent and concentration-dependent manner.②2.5×10-1mol/L sodium acetate and 1×10-2mol/L sodium caprylate induced cell cycle S arrest of MDA-MB-231 cells. 5×10-2mol/L induced cell cycle G2/M arrest of MDA-MB-231 cells. 1×10-2mol/L sodium butyrate induced cell cycle G0/G1 arrest of MDA-MB-231 cells. The apoptosis rate were 3.68% and 6.95% for 2.5×10-1mol/L sodium acetate and 1×10-2mol/L sodium butyrate at 72h.③MDA-MB-231 cells show the morphology of apoptosis under light microscope and electron microscope, when the cells was treated with 2.5×10-1mol/L sodium acetate, 1×10-2mol/L sodium butyrate and 1×10-2mol/L sodium caprylate. The apoptosis is not showed for 5×10-2mol/L sodium caproate. At the same time, the cells showed little death for 2.5×10-1mol/L sodium acetate and 1×10-2mol/L sodium caprylate.④Treatment of MDA-MB-231 cells with 2.5×10-1 mol/L sodium acetate, 1×10-2mol/L sodium butyrate and 1×10-2mol/L sodium caprylate induced lower levels of apoptosis, in which ladder-shaped stripe are shown in the DNA gel electrophoresis. The order induced apoptosis is sodium butyrate, sodium acetate, sodium caprylate and sodium caproate.⑤The Bcl-2 protein level of MDA-MB-231 cells was significantly decreased for 1×10-2mol/L sodium butyrate and 1×10-2mol/L sodium caprylate, the bcl-2 protein level of MDA-MB-231 cells was not influenced for 2.5×10-1mol/L sodium acetate and 5×10-2mol/L sodium caproate .CONCLUSIONS:①sodium acetate, sodium butyrate, sodium caproate and...
