Objective:To investigate effects of sodium butyrate(NaB) on the proliferation of human colon cancer cell line SW620, and to explore its possible mechanisms.Methods:In vitro, the SW620 cells were cultivated in RPMI1640 medium,which contain 10% calf blood serum and were placed in a constant temperature oven of 37℃and 5% CO2,then treated with different concentrations sodium butyrate (0,1.0,2.0,4.0,10.0mmol/L) and cultured for 24,48,72h respectively on the logarithmic phase of growth.Cell morphology changes were observed under inverted phase contrast microscopy;the growth inhibition was detected by MTT assay.The distribution of cell cycle was detected by flow cytometry(FCM).The expression of cyclin-dependent kinase inhibitor p21WAF1mRNA & VEGFmRNA was detected by reverse transcription- polymerase chain reaction(RT-PCR),the protein expression of p21 WAF1&VEGF was analyzed by Western Blot.Results:1.After treated by sodium butyrate,in comparise with control group, cells revealed obvious changes.2.Compared with the control group, the proliferation rates of SW620 cells treated by different concentrations(1.0,2.0,4.0,10.0 mmol/L)of sodium butyrate were inhibitied significantly. The inhibitory effect of NaB on cell proliferation was in a time-and-dose dependent manner.3.After incubation with2.0,4.0,10.0mmol/L NaB for 24h,the cell inhibitory rate was gradually increased, the ratio of S-phase cells decreased from 28.99%to 14.39%, 6.54%, 5.04%,respectively(p<0.05), meanwhile, the ratio of G0/G1-phase increased to 65.71%,78.98%,90.40%(p<0.05).4.The expression of p21WAF1 was upregulated by sodium butyrate at 24h both in mRNA and protein level,VEGF was downregulated,as the treatment concentration increased, the effects trended to be strengthened.Conclusion:Sodium butyrate can inhibit the proliferation of human colon cancer cell line SW620 and inhibitory rate in a time-and-dose dependent manner,and arrest of the SW620 cells in G0/G1 phase.Maybe,its mechanism was associated with upregulation of the expression of p21WAF1& downregulation of the expression of VEGF.
