Background and objectiveHepatic ischemia-reperfusion injury (HIRI) is the main problem during the operation of hepatolobectomy or liver transplantation. It is the prime reason of liver function failure after liver operation. Now the mechanisms of HIRI are considering: microcirculatory disturbance;oxygen free radical;calcium paradox;cellular factor and inflammatory factor, et al. The oxygen free radical may be the most important factor above all.The Acanthopanax senticosus is tranditional Chinese medicine, and has versitile bioactivities.It has been proved to be have good effects on ischemia-reperfusion injury on other diseases. We want to study the protective effects of acanthopanax senticosus injection against hepatic ischemia injury on liver function and oxygen free radicalMaterial and MethodsTwenty rabbits was random divided into two groups. Each was made a model of hepatic ischemia-reperfusion injury: operation of blocking the hepatic portal 45 minutes for hepatic ischemia, then recovering the blood supply 45minutes for reperfusion. One group received 5ml acanthopanax senticosus injection intravenous injection into portal vein system before the hepatic portal blocking. The another group received the same dose of normal saline. Blood ALT, AST, LDH, C-reactive protein, caeruloplasmin and transferrin were tested to observe the liver function changing. Tissue MDA, SOD were tested to observe the oxygen free radical deleting. The microscope and electron microscope for comparative pathologic changing were made too.Result30 minutes after portal blocking, acanthopanax senticosus group seral ALT 393.7 ±78.4U/L, AST 1444.4±194.8U/L, LDH 2496.0± 315.5U/L, liver tissue MDA 0.849 ± 0.085nmol/mgprot. SOD 110.1 + 10.8NU/mgprot;contral group seral ALT519.1+67.7U/L, AST1731.9+163.2U/L, LDH 3657.7+ 374.0U/L, liver tissue MDA 1.190 + 0.170nmol/mgprot, SOD 92.6±9.5NU/mgprot, PO.01. 45 minutes after recovering, acanthopanax senticosus group serai ALT 1453.4 +98.7U/L. AST 1734.9+228.4U/L, LDH 3159.2+263.4U/L, MDA 1.196 + 0.154nmol/mgprot. SOD 93.1±10.2NU/mgprot. CRP4.474 +0.292 mg/ L, caeruloplasmin 150.6 + 9.0 mg/ L;contral group serai ALT 2500.4+ 334.6U/L , AST 2522.2 + 259.7U/L . LDH 4297.8±313.6U/L . liver tissue MDA 1.680 + 0.139nmol/mgprot . SOD 70.5 +8.2NU/mgprot, CRP 5.715 + 0.425 mg/ L, caeruloplasmin 176.0 + 9.3 mg/ L, P<0.01.The pathologic changing was found that the hepatic lobules of the acanthopanax senticosus group was better than the contral group , and the degree of the cellular edema of the acanthopanax senticosus group was less than the contral group at the time of 45 minutes after recovering.Conclusion1. The oxygen free radical attends the progress of hepatic ischemia-reperfusion injury.2. The acanthopanax senticosus can protect against hepatic ischemia injury by protecting the cellular membrane, eradication of free oxygen and conserving the synthensis function of hepatocell.
