ObjectiveCombing the method of histopathology and technology of confocal microscope, the change of corneal morphology of rabbit using corticosteroid or NSAIDS was observed after LASIK , to understand the effect of drug to corneal wound healing after LASIK. The test will provide theoretical basis to choose correct medicine to reduce complication.Methods1, LASIK was performed in 18 rabbits both eyes, and the rabbits were dropped by tobramycin, fourth a day ,three days .2, Choose 12 rabbits at random after LASIK, drop corticosteroid (0.1%FML, experimental groupl)at right eye, and NASIDs at left eye(experimental group2).both groups were dropped 3,2,1 every day and dappered gradually 1 times every week. Other rabbits were dropped nothing (control group).3, 4 rabbits of experimental group and 2 rabbits of control group were examined by confocal microscope at 3days,lmonths,3months,after LASIK, then were sacrificed and acquired cornea to perform light microscope and electron microscope respectively.Results1, The corneal epithelial thickness begin increasing post-operatively, and is highest at 1 months ,the change of the corneal epithelial thickness of bothexperimental groups were smaller than control group .The differences were statistically significant.2^ Corneal flap thickness were no statistically differences at every interval , but the change trend is same to one of the corneal epithelial.3% The cell count in anterior stroma was lower post-operatively, The change number of anterior stroma cell of experimental groups was higher than control group, the differences were statistically significant.4> The cell count in posterior stroma were observed no remarkable change post-operatively.5^ Confocal microscope : corticosteroid may damage epithelial. But corticosteroid can reduce the secretion of ECM. Corneal epithelial change were no obvious differences between experimental group2 and control group.6% Light microscope : the proliferation of epithelial of control group was more evident than experimental groups. At early stage after LASIK. The arrange of collagen fiber of control group was more disorder than experimental groups. At 3 months, there was no differences among the 3 groups .7n Electron microscope: the epithelial surface of experimental group 1 appeared apoptosis, unhealthy epithelial cell. The finger protrude structure decreased obviously. Cytoplasm of epithelial cell was loose. The expression of experimental group 2 was weaker than experimental group 1.Conclusions1 -. Corticosteroid can inhibit the epithelial proliferation of cornea after LASIK . The function of NSAIDs was weaker than corticosteroid.2> Corticosteroid had epithelial toxicity to cornea. NSAIDs had the lighter effect to corneal epithelial.3> Corticosteroid can inhibit the proliferation of ECM of cornea and anterior stroma collagen fiber. The effect of NSAIDs was weaker.4> There were pros and cons of corneal wound healing between Corticosteroid and NSAIDs. According to the different condition to choose different drug , may reduce the incidence of regression and complication after LAS IK.
