Objective: To observe the activation of NF-κB in rats SAP model and the treatment effects of inhibitor of NF-κB , Chinese medicine glucocorticoid and octreotide Methods: Rats SAP model were established by intracholedochal injection of sodium cholate sulfur, then were treated respectively with glucocorticoid, pyrrolidine dithiocarbamate(PDTC), octreotide or LaiFuChengQiTang. pancreatic tissue of one half of the rats were harvested 3 hours after the operation for NF-kB examination;the blood samples of anather half of the rats were collected 24 hrs after the operation, for the measurement of amylase and endotoxin, and patho-diagonosis. Results: The levels of amylase, Endotoxin and activation of NF-kB in SAP model group increased remarkably, Compared with control group;And the pathologic changes accorded with acute hemorrhagic and necrotic pancreatitis. The mortality and pathologic damage reduced both in PDTC and herbs groups, and the level of amylase, endotoxin and activation depressing of NF-κB also be observed in the same time. Glucocorticoid showed nosuch effect and Octreotide had distinct effect on reducing amylase level and endotoxin , but no effect on activation of NF-kB. Conclusion: PDTC and LaiFuChengQiTang has assured effect on SAP by dicreasing endotoximia and depressing the activation of NF-kB.
