ObjectiveBy detecting the different level of p21ras and CD34 in 60 paraffin - embedded tumor tissue of human colorectal carcinoma by immuohistochemistry, we will analyse their clinicopathological value and relationship. Further to approach the relationship of signal - transduction pathways with tumor angiogenesis in human colorectal carcinoma.Method1. Specimens: a group of paraffin - embedded tumor tissue of colorectal carcinoma patients from oncology department of the first clinical hospital of China Medical university were used.2. All the specimens were investigated by HE. The expression of p21ras, CD34 was investigated by immuohistochemistry.3. Statistics analysis : SPSS10.0 for windows was used. T - test and chi -squared test were the detail ways.ResultsIn 60 colorectal carcinoma, the positive rate of p21ras was 70% , MVD was 55.76 ±17.47. The expression of p21ras,CD34 was no significant difference in different gender ,histological type. MVD with p21ras positive expression was sig-nificantly higher than negative. The expression of p21ras was significantly correlated with tumor size ^lymph node metastasis and depth of invasion. MVD was not correlated with tumor size, was significantly correlated with lymph node metastasis and depth of invasion.DisussionsOncogenesis is a complex procedure. The growth of solid tumor depends on continuous angiogenesis. The view that tumor progression and metastasis depends on angiogenesis was prefered by Dr. Folkman earlist in 1971. Then more and more studies about tumor angiogenesis were taken on, angiogenesis becomes the hot spot of tumor research day by day. The point that angiogenesis is correlated with many kinds of cytokines , gene and gene production has been accepted by most.Currently, the relationship of signal - transduction pathways with tumor ari-giogenesis becomes a new research area. Three distinct MAPK pathways have been expounded: (1) JNK/SAPKs pathway;(2 ) p38 pathway;( 3 ) ERK pathway. ERK pathways was the one that human know the most clearly. The Ras — Raf - MEK - ERK (ERK) pathway plays a critical role in many aspects of cel-luar function. The difference of ERK pathway with the two others was that ERK pathway inhibits apoptpsis, however the two others stimulate and promote ap-optpsis. But ERK pathway induces apoptpsis was found in some studies. The activation of ERK pathway was always found in human carcinoma, it is due to so many agents, mutational activation of oncogene ras was one of those. It has been reported that ERK pathway could regulate the production of key angiogenic factors such as VEGFO Wilhelm SM prefer that Ras/Raf signal - transduction pathway was an important intermediary agent to the proliferation of tumor cell and angiogenesis. Liang JT prefer that mutation of K - ras was intimately corelated with tumor angiogenesis.Conclusions1. The expression of p21ras was significantly correlated with tumor size A lymph node metastasis and depth of invasion. MVD was significantly correlated with lymph node metastasis and depth of invasion.2. MVD with p21ras positive expression was significantly higher than negative.3. Mutation of oncogene ras may enhance angiogenesis in human colorectal carcinoma, further to approach the the relationship of signal - transduction pathways with tumor angiogenesis.
