Survivin Antisense Oligonucleotide Induces Gastric Carcinoma Cells Apoptosis And Sensitizes The Cells To Resveratrol

Objective: To explore the expression of survivin in gastric carcinoma cell SGC7901 and the effect of antisense oligodeoxynucleotides (ASODN) of survivin gene on apoptosis and resveratrol sensitivity of gastric carcinoma cells.Methods: Our study is based on cell-culture in-vitro and anti-survivin phosphorothioate ASODN was synthesized and transfected into gastric carcinoma cells by lipofectin. MTT assay was used to detect sytotoxicity. Apoptosis was observed by fluorescence microscopy and Transmission electron microscope and flow cytometry. Survivin expression was determined by RT-PCR and immunohisto-chemistry.Results: ①Expression of survivin is high in gastric carcinoma cells, ②survivin ASODN inhibited the cells proliferation in a dose and time dependent manner. ③ A higher apoptosis rate (32.5%) could be induced in gastric carcinoma cells by survivin ASODN as compared with that induced by the sense oligodeoxynucleotide (11.8%)(P<0.05). ④ The expression of survivin mRNA and protein significantly decreased after treatment with survivin ASODN . ⑤There was a significant increase or cell inhibition rate after exposuring to the combination of survivin ASODN and resveratrol as compare to resveratrol or survivin ASODN alone (Both P<0.05).Conclusion: Taken together, our suggest that survivin are important survivalfactor for gastric carcinoma cells .Survivin ASODN is able to inhibit the proliferation of gastric carcinoma cells, induce the apoptosis, and enhance the sensitivity of gastric carcinoma cell line SGC7901 to resveratrol via specific down regulation of survivin expression.
