In this thesis, the chemical constituents in Cortex Moutan were studied by high performance liquid chromatography coupled with mass spectrometry (HPLC/MS) and nuclear magnetic resonance(NMR). The quantity of Cortex Moutan was evaluated by qualitative and quantitative analysis. Focused on the problem of quality control (QC) of Chinese Medicine preparation, and its application of shuangdan granules were carried out. The main contents and productions were as followed:1. Study on the quality evaluation and chemical components of Cortex Moutan. Five key pharmacological compounds, namely gallic acid, oxypaeoniflorin, paeoniflorin, benzoylpaeoniflorin and paeonol, were determined by HPLC/UV. Simultaneously, the chromatographic fingerprints of Cortex Moutan were established. Nineteen common peaks were confirmed to reflect the phytoequivalence generally, and all the peaks were identified by high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry(HPLC/ESI-MS~n). Eight monoterpenoids were isolated and identified by HPLC/MS, MPLC, prep-HPLC and NMR rapidly. This quality evaluation method which combined the chromatographic fingerprints and quantification assay to ensured that the phytoequivalence and pharmacological effects of Cortex Moutan and was successfully applied to the quality control of Cortex Moutan.2. The quality analysis of shuangdan granules was studied. Develop a HPLC method for the determination of salvianolic acid B and paeoniflorin in Shuangdan granules. The method was accurate, reproducible, reliable, and could be used for quantitative control of Shuangdan granules.
