Chinese Medicines are playing an important role due to their effectiveness in Chinese public life for thousands of years. However, the quality control of Chinese Medicines is a difficult mission because of the complex chemical constituents and the ambiguous pharmacological components. To reveal the chemical and pharmacological efficacy substance of Chinese Medicines is the key to construct the standardization of quality evaluation of Chinese Medicines, propel the development of the Chinese Medicines science and the modernization of the Chinese Medicines industry. In this thesis, the high performance liquid chromatography tandem mass spectrometry (HPLC-MS) based methodologies of studies on chemical substance of "SHUANGDAN" formula and the variation of the chemical constituents of compatibility of Radix Salviae Miltiorrhizae and Cortex Moutan were constructed. The main contents and productions were as followed:1. An HPLC-DAD-ESI/MS" method was developed for simultaneous analysis of the chemical constituents in "SHUANGDAN" granules, which would facilitate the improvement of its quality control standards. In this chapter, fragmentation characters of 10 reference compounds, including phenolic acids, diterpenoid quinones and monoterpenoids were investigated and fragmentation patterns were proposed. Then according to the retention time, characteristic UV absorption profile, the information of molecular weight and structure provided by ESI/MS", 28 constituents including 14 phenolic acids, 6 diterpenoid quinones, 6 monoterpenoids and 2 other components were identified, while some isomers were distinguished based on the MS~n spectra. This method was rapid and reliable for identification of constituents in complex chemical system, and the newly fragmentation patterns proposed could be extended to the compounds elucidation with similar framework.2. The chemical constituents of compatibility of Radix Salviae Miltiorrhizae and Cortex Moutan were studied by HPLC-MS. Nineteen constituents were identified and attributed, among them eleven from Radix Salviae Miltiorrhizae and eight fromCortex Moutan. Comparing to the peak areas of constituents, it is indicated that the content of phenolic acids and monoterpenoids in combined-decoction was higher than those in single-decoction, but the content of tannins was lower.The results illuminated that the HPLC-MS technique is a powerful tool for the study of chemical substance of Chinese Medicines, which also could be applied for optimization of their quality control methods.
