Guanbaifu is the tuber root of Aconitum coreanum (Levl.)Rapaics, which is a kind of Chinese traditional herb and widelydistribute in Heilongjiang, Jilin and Liaoning Provinces.According to the references, there are a lot of dissertationsabout some compounds, such as steroids, diterpene alkaloids,hypaconitins etc. from the tuber root of A. coreanum (Levl.)Rapaics were isolated and identified previously. Modernpharmacological studies reported that this medicinal herb had theeffects of analgesia, antiplatelet aggregation, anti-inflammatory,anti-arrhythmis etc. At the same time the Guanbaifu base G,Guanbaifu base A and Guanbaifu base I were confirmed as themain bioactivities constituents in this plant. But there are only afew papers reported about the chemical consitituents andbioactivities of the leaf and stem of A. coreanum (Levl.) Rapaics.The chemical constituents and its bioactivities of the leaf andstem of A. coreanum (Levl.) Rapaics were focused on this thesis.5kg leaf and stem of A. coreanum (Levl.) Rapaics collected fromJingyu county, Jilin province were extracted with 85% ethanol at50℃ for 24 h firstly, the ethanol extract were obtained byreclaiming away the ethanol solvent. The ethanol extract aqueoussolution was acidified with 1% HCl, and then got rid of thedeposits by filtration method, after that the solution which wasadjusted pH to 9-10 by adding ammonia was re-extracted withchloroform. From the chloroform parts, 8 compounds wereobtained by way of silica gel chromatography, TLC and HPLC etc.Among them 5 compounds' structures were elucidated byphysico-chemical properties and spectroscopic evidences (MS, IR,1HNMR, 13CNMR, 1H-1Hcosy, HMBC, DEPT) respectively. Theyare Guan-fu base I(I),Guan-fu base R(Ⅱ),β-Sitosterol(Ⅲ),D-mannitol(Ⅳ),and Daucosterol(Ⅴ), in which the compound (Ⅱ)is a new diterpene alkaloid.The pharmacological activity of the ethanol extract from theleaf and stem of A. coreanum (Levl.) Rapaics was studies. Theresults showed that it has the effects of inhibiting metalloproteinaseactivity.
