objective: study expression of the peritoneal noxious stimulation making c-fos and NOS in central nervous system. Method: 1% acetic acid is injected into peritoneal cavity of rat and making the abdomino-pain model. Choose 56 healthy Wistar rats, which randomly are divided experiment groups(n=48) and controlling group(n=8) (A group ) v/hen male and female sex is not limited. According experimental requirement, the animals of experiment group survive respectively 30minutes(n=8) lhour(n=8) 2hours(n=8) 4hours(n=8) , 6hours(n=8) 8hours(n=8) and are divided into B group C group D group E group F group and G group after 1% acetic acid is injected into peritoneal cavities of rats. Then respectively draw the materials from rats ,and section. The localization of c-fos and NOS positive neurons in central nervous system was detected by means of immunohistochemistry (ABC method). Result: i% acetic acidis injected into peritoneal cavity,and Rats are obviously irritable when they are conscious, and back bones of rats arche. After animals are irrigated and fixed, anatomical peritoneal cavities of rats have exposed peritoneal engorge merit, dropsy and exudation of greater omentum. Fos of A group is little expressed in central nervous system. Experiment groups compared with controlling group, the result has statistical value(p<0.05) . the fos positive neurons are also distributed in both sides of CNS, located within T6-L3 posterior horn of spinalcord nucleus of solitary tract parabrachial nucleus locus ceruleus nucleus . dorsal nucleus of median suture hypoth-alamus ; paraventricular nucleus supraoptic nucleus amygdaloid nucleus. Experiment groups compared with controlling group; after 1% acetic acid is injected into peritonea! cavity of rat; the positive cell number of NOS obviously increases all at 1hour 2hours 4hours 6hours in above mentioned nucleuses (P<0;01) . Compare with c-fos; the expression of NOS peaks at 1hour; NOS expresses Ihour earlier in peak than c-fos. Conclusion: NO conducts c-fos expression which is caused by nociceptive stimulation; and is necessary condition which further induces c-fos expression in the cell. Above mentioned nucleuses might involve in rat abdomino-pain which is caused by peritoneal noxious stimulation.
