Objective: To investigate the effect of hypertonic saline (HTS) on expression of TNF-α, IL-10mRNA in pancreatic and lung tissue of rats with severe acute pancreatitis(SAP). Methods: SAP was induced in rats by intraperitoneal injection of 20 per cent L-Arginine twice at an interval of 1 hour. Animals were randomized into four groups(each n=12): (1)Controls;(2)SAP without resuscitation;(3) SAP plus intrarenous resuscitation with normal saline (NS, 0.9 per cent sodium chloride 2ml/kg); (4) SAP plus intrarenous resuscitation with HTS (7.5 per cent sodium chloride 2ml/kg) at 0, 12,24,36h. Pancreatic and lung tissue were collected at the 24h and 48h after induction .The expression of TNF-α IL-10mRNA in pancreas and lung was detected by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) technique. Pathologic study of pancreas was also performed. Results: Compared with those resuscitation with NS,the expression of TNF-α mRNA of the animals received HTS decreased significantly (p< 0.05), and the expression of IL-10mRNA increased more significantly after the induction of SAP (P< 0.01).At the same time ,pancreatic and lung histological scores were reduced in animals treated with HTS (P< 0.01).Conclusions : Hypertonic saline resuscitation result in an effective method to restore the balance of pro-and counter-inflammatory cytokines.
