Objective In order to observe the renoprotective effect of valsartan and examine the expression of connective tissue growth factor (CTGF) in the kidney of spontaneously hypertensive rats.Methods The spontaneously hypertensive rats(SHR) were randomly assigned to control group and valsartan treatment group ,valsartan treatment group was gorged with valsartan 30mg.kg-1. d-1. Wistar-Kyoto Rats(WKY) served as normal control,of which blood pressure, urinary β2-microglobulin, Bun , and Scr were acquired at 12-week old and 24-week old respectively, at the end of experiment, the renal expression of CTGF were determined.Results 12 weeks later, In the SHR control group,the blood pressure, urinary β2- MG,except Bun, and Scr were significantly increased compared with the other two groups(p<0.05 > p<0.05 ) .The change of urinary β2-MG : The lever of urinary β2-MG has no significantly difference at different time in the WKY group. the lever of it in the SHR control group and the SHR treat group has significantly changed than before experiment (p<0.01, p<0.05),It was much higher in the SHR control than that in other two groups after experiment(p<0.01, p<0.01) ,And it has obviously decreased in SHR treatment group than before experiment(p<0.05).Semi-quantitative assessment of the immune-histochemical staining showed that :Renal expression of CTGF in SHR control group were significantly increased compared to that in WKY group and treatment group(p<0.01,p<0.01) . At the same time, The expression of CTGF was also found positively arrelated with the increasing of urinary β2-MG(p<0.05).Conclusions Valsartan not only significantly reduced the extent of proteinurina of SHR, but also decreased the expression of CTGF in the kidney of spontaneouslyhypertensive rats .CTGF may be involved in progress of this renal disease.
