Objective: Through the observation of YangGanYiShui Granule on12week old SHRrenal function, the expression of renal cortical STAT1, p38MAPK, Wnt proteinexpression and renal cortex of α-SMA, CTGF mRNA and protein expression of mRNA,CTGF in renal cortex of the relationship between the expression of α-SMA in renalcortex, and STAT1,p38MAPK, Wnt protein, the relationship between the expression ofMA, β2-MG, urine the content of NAG and STAT1in renal cortex, p38MAPK, Wntprotein and renal cortex of α-SMA, CTGF mRNA and protein, and the interventioneffect of YangGanYiShui Granule on the early renal damage of hypertension, to explorethe pathogenesis and YangGanYiShui granule renoprotective mechanism from themolecular level, in order to deepen and enrich the theory connotation of traditionalChinese medicine.Methods: The purchase of12week old spontaneously hypertensive rats from BeijingWeitong Lihua Experimental Animal Technology Co. Ltd.(SHR)40and Wistar-Kyoto(WKY)10rats, fed for1weeks, weighing, marking the lifetime test by picric acid, SHRwere randomly divided into4groups, namely YangGanYiShui granule high, low dosetreatment Bela Pury group, control group, model group. Observation of12week oldSHR urinary MA, urinary β2-MG, NAG and serum Scr, BUN content; to observe theexpression of α-SMA, renal cortex STAT1, p38MAPK, Wnt, CTGF protein expressionand renal cortex of α-SMA, CTGF mRNA; observation of YangGanYiShui Granule onSHR urinary MA, urinary β2-MG, NAG, expression renal cortical α-SMA, STAT1,p38MAPK, Wnt, CTGF protein expression and renal cortex of α-SMA, CTGF mRNA,and the relationship between the expression of renal cortical CTGF mRNA and proteinexpression and renal cortex STAT1, p38MAPK, Wnt protein, expression of urinary MA,β2-MG, NAG content and renal cortex STAT1, p38MAPK, Wnt protein and renalcortex of α-SMA, CTGF mRNA and protein affect the relationship between hypertension; early renal damage mechanism and YangGanYiShui granule control waysand links.Results: First, General condition of rats (colour, mood, activity, diet, etc.), aftertreatment, YGYSG general improvement evident in the low dose group approachedWKY group rats;YGYSG high-dose group as the growth of the drug intervention timetearing and fight decreased gradually, response insensitive; That, compared with modelgroup, benazepril group did not significantly improve performance status. Second,Model group increased with the increase of age of rat blood pressure, urinary MA, urinebeta2-MG, NAG were significantly higher in the WKY group (p<0.01); Treatmentbefore and after model group SHR rats blood Scr, BUN, rats was significantly higherthan that of WKY group (P<0.01); After treatment the treatment group can significantlyreduce SHR rats blood Scr, BUN (P<0.01), but compared with no significant difference(P>0.05). Third, SHR after treatment groups of CTGF in the kidney tissues were higherthan that of WKY group (P<0.05or<0.01), the group that split renal cortex CTGFexpression quantity decreased significantly in the model group (P<0.01); YGYSG inhigh dose group of renal cortical CTGF expression is less in the model group (P<0.01),YGYSG in high dose group of renal cortical CTGF expression with Mr Comparisonbetween the captopril group, with statistical significance (P<0.05); YGYSG in low dosegroup of renal cortical CTGF expression compared with Mr The captopril group, therewas no statistically significant difference (P>0.05).Conclusion: YGYSG can improve SHR rats general situation,CTGF inducedspontaneously hypertensive kidney ECM accumulation exist multiple relatedtransduction path; YGYSG by intervening CTGF induced renal parenchymal cells toexpress alpha a-smooth muscle actin muscle fiber cell transdifferentiation and reduceurinary MA, β2-MG, NAG, blood content, protect the kidney.
