In order to determine the biomarkers in differential diagnosis between hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (ICC),we used laser capture microdissection coupled with two-dimensional gel electrophoresis and electrospray ionisation tandem mass spectrometry to discuss the differential expression of proteins between them.78 protein spots were selected and 28 significant proteins were identified from hepatocellular carcinoma and intrahepatic cholangiocarcinoma tissues,20 proteins and 8 proteins only expressed or upregulated in hepatocellular carcinoma and intrahepatic cholangiocarcinoma tissues each.Immunohistochemical method showed that BK125H2.1 expressed in nomal tissue and 92.5%HCC tissue,5 % ICC tissue showed immunoreactivity for BK125H2.1,so laser capture microdissection is a key technologic method in proteomic study of cancer tissue,which can get rid of irrelevant tissue effectively.The proteins revealed in this experiment such as BK125H2.1 may serve as novel candidated biomarker for differential diagnosis of hepatocellular carcinoma from intrahepatic cholangiocarcinoma.
