By detecting the survival and differentiation of bone marrow mononuclear cells (MBMCs ) transplanted into myocardial infarction (MI ) region and the effect of cardiac function , this study try to discuss the efficacy and security of MBMCs transplantation in treatment of MI.Methods: (1) Myocardial infarction was induced by ligation of left anterior descending coronary artery . after two weeks, the MBMCs were injected into the peri-infarction myocardium. Four weeks after MBMCstransplantation, the examination of echocardiography , haemodynamics , immunohistochemistry , confocal laser scanning micrography and transmission electron micrography was performed in all groups to observe the changes of left ventricular function and the efficiency and security of MBMCs transplantation. (2) MBMCS were transfused into re-open coronary vessel from embolism through microcatheter . cardiac function and MI area were examined by echocardiography and myocardium radioisotope scanning, cardiac blood pool development, then we evaluate the security and validity of MBMCS transfusion treatment.Results: (1)Four weeks after MBMCs transplantation, the hearts with transplantated cells had better cardiac function than the control hearts did(P<0.05). Six weeks after MBMCs transplantation, the eximanations of immunohistochemistry and transmission electron micrography were performed, and the result showed that the transplantated cells had been survived and proliferated in the transplanted area . (2) The left ventricular systolic and diastolic function was improved among the patients received MBMCS transplantation, the reduction of MI area and the increase of ratio of myocardium nuclide ingest were observed in the same group.Conclusion: These results demonstrate that transplantation of MBMCs can improve cardiac function. The primary clinical treatment demonstrate that self MBMCS transplantation is secure and valid.
