To explore the dedifferention and its induction in glial cells and nerve regeneration by autograft pre-degenerateded nerve, changes of morphology and expression of proteins and genes were studied by immunohistochemistry ABC method, in situ hybridization histochemistry, double immunofluoresence histochemistry and gene chip after optic nerve injury in rats. After optic nerve injury, expression of Nestin was up-regulated. Gene expression in macroglia cells changed and so on. Expression of Nestin was further up-regulated by autograft perpheral nerve. In addition, autograft peripheral nerve regulated expression of a lot of genes in macroglia cells. These results suggest that there be not a successful regeneration, expression of genes be regulated in macroglias, Nestin expression increase, and macroglia cells dedifferentiate after optic nerve injury. Autograft peripheral nerve can weaken the hibitory environment caused by optic nerve injury and promote the optic nerve regeneration. Autograft peripheral nerves can regulate gene expressions in macroglia cells and induce the dedifferentiation of macroglia cells in the optic nerve.
