Objective: To study the effects of intrastriatal transplantation of BMSCs on the promotion of functional recovery after stroke in rats.Methods: The BMSCs was separated, purificated, proliferated and preinduced by FGF-2 in vitro. BrdU were transfected into BMSCs before trangplantation. Audult male Wistar rats were subjected to tMCAO and received grafts into the striatums at 7 days after stroke, rats were divided randomly into 5 groups: control groups: (1) sham-operated group, n=7: preinduced BMSCs injected into the nomal striatum;(2)stroke control group, n=7: MCAO alone;(3 )sham-grafted group, n=7: rats received equivalent volumes of saline. Experiment groups: (4) n= 10: injection of BMSCs preinduced by FGF-2;(5) n=10: transplantation of BMSCs.Behavioral tests were performed before, 7, 14, 21and 28 days after transplantation respectively. Rats were killed at 28 days after transplantation. The electron microscope examination was performed. The pathologic examination was also performed and secretion of neural growth factor BDNF was measured. Results: Singnificant recovery of Neurological Severity Score were found in rats in experiment groups at 28 days after transplantation compared with sham-grafted group.Group4 is better than group5. Single and double immunohistochemical staining was performed. There were BrdU-reactive cells within the brain tissue of group 1 , 4, 5. More cells of BrdU -labeled can be seen in group 4, but none of Brd U-reactive cells expressed NSE and GFAP. Thesecretion of neural growth factor BDNF in experiment groups was much more than the sham-grafted experiment group.Group4 is higher than group5. The electron microscope examination was performed. Most of transplanted BMSCs have survived and contact with host cells .Some have features of differentiation. Conclusion: The current findings suggest that the intrastriatal transplanted BMSCs survived in the ischemic brain and improved functional recovery of rats after stroke .Functinal recovery from NSS tests was significantly improved in the rats receiving preinduced BMSCs compared with BMSCs alone.
