| Object To test and compare the antiviral effects of lemon extracts 0# and 3# with different experimental methods on Newcastle disease virus and Coxsackievirus B3;To test and compare the antibacterial effects of lemon extracts 0# and 3# on Escherichia coliform (E.coli) , Staphylococcus hemolytic (S.hemolytic) , Pseudomonas aeruginosa (P.aeruginosa) and Candida albicans.Method Direct killing effect: To mix Newcastle disease virus andCoxsackievirus B3 with drug 0# and 3# in vitro and the four pairs react for 1 hour, then drop them into vero cells and react for 1 hour,then throw the fluid away and add maintain-fluid into the vero-cells and culture for 72 hours;Preventive effect: Drop drug 0# and 3# with different concentrations into Vero cells for 24 hours , then drop the Newcastle disease virus and Coxsackievirus B3 into the cells instead of the drug only,and react for 1 houe, then add maintain-fluid instead of the mixtures,then culture for 72 hours;Cure effect: To drop the Newcastle disease virus and Coxsackievirus B3 into Vero-cells for 1 hour,then drop 0# and 3# into Vero cells instead of virus ,and culture for 72 hours. Compare the effects of the three methods separately with the effect of the virus only on the cells according to the CPE. Prepare the fluid with the activated bacterial and one fungus,to count the number of the three bacteria and one fungus . Drop 0.2ml bacterial fluid separately into the the plates with culture medium, spraid the fluid and lay the plates on the operative table for 20 minutes. Divide drug 0# and 3# with different concentrations,and drop 10μl intopieces of filter paper separately,then lay them on the plates and interval 18mm between each two pieces. Then culture for 24h to 48h at the temperature 36°C.Result In the experiment of the direct killing effect of Lemon extracts 0* onNewcastle disease virus,experimental group has obvious difference with control group(p<0.05);in the experiment of the preventive effect of the Lemon extracts 3* on the Coxsackievirus B3, experimental group has obvious difference with control group(p<0.05);in the experiment of the cure effect of the Lemon extracts 0* and 3* on the Newcastle disease virus and Coxsackievirus B3, experimental group has no obvious difference with control group(p>0.05). Lemon extracts 0* can restrain three bacterial and one . Lemon extracts 3* can restrain E.coli and Pa. But no effect on S.hemolytic and Candida albicans.Conclusion Lemon extracts 0* has direct killing effect on Newcastle diseasevirus;lemon extracts 3* has preventive effect on Coxsackievirus B3;Lemon extracts 0* and 3* have no cure effect on Newcastle disease virus and Coxsackievirus B3. Lemon extracts 0* can restrain three bacterial and one , especially on staphyiococcus hemolytic. Lemon extracts 3* can restrain E.coli and Pa, especially on Pa is more powerful.but no effect on S.hemolytic and Candida albicans.
