| ObjectiveTo explore a new method of chemotherapy for orbital malignant tumors. It can increase the drug concentration around the tumor and reduce systemic side effects by injecting cytoreductive drug into lesions. This research aim at improving cure rate and reducing occurrence of orbital exenteration. This experiment offered an animal foundation and clinical reference for locally administered treatment for orbital malignant tumors. MethodsOrbital adenoid cystic carcinoma (ACC) was selected for the research.1. The drug sensitivity of ACC in vitro was determined by MTT assay to screen the ACC special sensitive chemotherapy medicine.2. Orthotopic nude mouse model of orbital ACC was built by injecting ACC cell suspension.3. The nude mice bearing cancer were divided into 5 groups. The growth curve of tumor was investigated. The effects of systemic chemotherapy and locally chemotherapy were compared. The most effective drug concentration was concluded by comparing 3 different drug concentration.4. The mechanism of VCR was investigated rudimently by comparing the electron microscope presentation of tumor before and after chemotherapy. The side effect of intralesion cytoreductive drug injection was studied by observing the local skin and parenchyma in the mice orbit.Results1. The sequence of the drug sensitivity of ACC in vitro was: VCR, ADM, PYM, DDP, 5-Fu, CTX. VCR was the special sensitive drug and the CTX was themost insensitivity one.2. The ACC model were succeeded in all nude mouse. The range of tumor bulk before chemotherapy was 17.20 mm3 to 49.32 mm3, the mean is 30.55mm3.3. The tumor inhibition rate of VCR2 u g> VCR3 u g, VCR4 ii g and systemic chemotherapy was 77.82%> 93.09%> 82.82% . 66.65% respectively. The difference between VCR3 u g and systemic chemotherapy exhibited statistical significant (PO.05).4. The histologic examination of skin and parenchyma in the mice orbit of local chemotherapy showed no pathologic changes.Conclusions1. Orthotopic nude mouse model of orbital ACC was built by injecting ACC cell suspension.2. VCR was the special sensitive chemotherapy medicine for ACC in vitro.3. It is an effective therapy for orbital ACC by inject cytoreductive drug into lesions. The best VCR concentration for intralesional injection was 0.03mg/ml.4. The pathologic results showed the method of intralesional injection of VCR is safe and effective. It would be one of effective method of chemotherapy for orbital ACC. |
