Immune Response Induced By Dendritic Cells Transfecting With Tumor Cells Total RNA Inhibits Growth Of Implanted Tumor In Vivo And Rejects Challenge Of Tumor Cells In Nude Mice

Objective: To investigate whether blood-derived DC transfecting with unfractionated RNA (total RNA) extracted from tumor cells of Adenoid cystic carcinoma can inhibits growth of implanted tumor in vivo and rejects challenge of tumor cells in nude mice.Methods: DCs were generated from Adenoid cystic carcinoma patients concentrated leukocyte fraction of blood. Peripheral blood mononuclear cells (PBMC) were further separated by density-gradient centrifugation and cells were resuspended and allowed plastic adherence. The adherent cell fraction was used for DC culture in CM supplemented with rhGM-CSF , rhIL-4 and rhTNF- α . After 5 days of culture, DCs were harvested. DC preparations fulfilling immature DC phenotypic criteria were used for subsequent RNA transfection. Total RNA was isolated from tumor cell line SACC83 of Adenoid cystic carcinoma and used to transfect the DCs. Transfected DC will irritate homogenic T lymphocyte to form cytotoxic T lymphocyte(CTL). Implanted the CTL to inhibits growth of implanted tumor in nudes and protect the nudes against the further challenge of SACC83 tumor cells.Results: The CTL induced by dendritic cells transfecting withtumor cells total RNA can protect the nudes against the challenge of SACC83 cells(experimental group, the incidence of the transplanted tumor was 22.2%,control group was 100%,P<0.05)and inhibits growth of implanted tumor in nudes(experimental group, 1293 ± 170mm2, control group, 669 +100mm2, P< 0 . 05).Conclusion :The immune response induced by DC not only inhibits growth of implanted tumor,but also protect against the challenge of SACC83 cells in nudes.The results suggested that,as a new concept of anti-tumor vaccine,DC transfecting with tumor cells total RNA of Adenoid cystic carcinoma may play an important role in treating and preventing this tumor.
