Expression And Significance Of Survivin In Carcinoma Of Prostate

IntroductionSurvivin , one member of IAP family , is considered as a kind of tumor specific apoptosis inhibitor. Prostatic carcinoma is one of the malignant tumors which have the highest morbidity in male in the west. But in recent years morbidity of prostatic carcinoma in China is increasing sharply and more attention are paid to it. Until now few studies were made on the relation between Survivin and prostatic carcinoma. In this study, we use immunohistochemistry method to detect Survivin expression in 33 cases of prostatic tumors to find its effect in carcinogenesis and progression of prostatic tumor.Materials and Methods1 Material;1. 1 Specimen: 33 cases of prostatic tumors and 26 cases of benign prostatic hyperplasia embedded in paraffin were obtained from the Department of pathology of the Shengjing Affiliated Hospital of China Medical University from 1990 to 2004.1.2 Main reagents: Survivin monoclonal antibody was obtained from Santa Cruz Company. SP staining kit, DAB Developer, Antibody Dilution were obtained from Beijing Zhongshan Biotechnology Company.2. Methods;Use SP immunohistochemistry method to detect expression of Survivin in prostatic tumors and benign prostatic hyperplasia tissue. Judgment Criterion:Buffy granules in cell membrane and kytoplasm is positive cell,no stain is negative. Judging by cancer cell staining degree, scope, distribution;negative ( - ) : positive cell(10%;light positive ( + ) : positive cell 10-25%;meddle positive ( ++ ) : positive cell 26% -50%;strong positive ( +++ );positive cell > 50%. Statistics method:x2 " test by SPSS12. 0 software.ResultsSurvivin protein expressing in cell membrane and kytoplasm is buffy granules;positive rate of Survivin protein expression in prostatic carcinoma is 78. 79% (26/33) ,in benign prostate tissue is 11.54% (3/26) , the difference is significant( P <0. 001) o Positive rate of Survivin protein expression in poorly differentiated and moderately differentiated cancer tissue is higher than well -differentiated cancer tissue the difference is significant (P <0. 05). difference of Survivin protein expression positive rate in poorly differentiated and moderately differentiated cancer tissue is also significant ( P <0. 05 ). Survivin protein expression has close relation with WHO Cancer clinical scale. In 33 cases of prostatic carcinoma in which Survivin protein expression is positive , the positive rate is 70.00% (14/20) in A - B stage ,92. 31 % (12/13) in C - Dstage. Difference of Survivin protein expression positive rate between clinical stages is significant (P<0.05).ConclusionExpression level of Survivin protein in prostatic carcinoma is higher than that in benign prostatic hyperplasia tissue. So Survivin may play an important part in carcinogenesis and progression of prostatic tumor. Expression level of Survivin protein has relationship with differentiation and staging of prostatic carcinoma: its Expression level increase with the decreasing of differentiation and the increasing of pathology stage. So Expression level of Survivin protein influence cancer malignant degree and progression and in some extent forecast prostatic carcinoma prognosis. Expression positive rate of Survivin in prostatic carcino-ma is 11.54% (3/26) and in prostatic carcinoma is 78.79% (26/33) , difference between the two teams is significant ( P <0. 001) ,so Survivin detection can distinguish them.
