Objective: Pass to establish an optimized project of ex vivo expansion and sub-lethally irradiated immunodeficiency mice of UCB transplantation, and to explore the effect of the engraftment capability and hematopoietic potential of the water-soluble chitosan with the umbilical cord blood transplantation (UCBT) in murine model. A promoted and choosed medicine of the biology device was established to transplant HSPC in murine model, and the significant mechanism of transplant UCB with the water-soluble chitosan was investigated in immunodeficiency mice.Methods: 1.The effect of different concentration of the water-soluble chitosan and cytokin (SCF+FL+IL-3) coordinated with different concentration of the water-soluble chitosan on proliferation of stem cells were detected by MTT and FCM. 2. Immunodeficiency mice were made by exposed to C ray. All mice were divided into 5 groups in random. A group: the normal mice;B group: model;C group: injecting 5×10~5 CD34+ cell umbilical cord blood (UCB) through the tail vein;D group: injecting 5×10~5 CD34~+ cell umbilical cord. blood(UCB)through the tail vein, while impregnating 500mg/d/kg.bw of the water-soluble chitosan;F group: impregnating 500mg/d/kg.bw of the water-soluble chitosan. Observe thelivability, the immunity resume and hematopoiesis rebuild of the immunodeficiency mice.. Result: 1. CS 5.0-20.0ug/ml stimulated the proliferation of UCB distinctly, lOug/ml of CS had better effects .When compared to the cytokine, CS coordinated with cytokine had better promoting effect. The proliferation multiple were 4.52±0.3649 and 11.88±1.2527, respectively. The difference between cell factor group and CS group was significant. Increase in concentration did not lead the increase in proliferation. 2. After transplant the livability of mice enhanced obviously, the immunity resume and hematopoietic reconstitution were all promoted. The effect was excellent when CS coordinated with the cytokine.Conclusion: 1. CS can stimulate the proliferation of UCB, when coordinated with the cytokine has synergistic effect. 2.The immunity resume and hematopoiesis rebuild can be promoted when transplant UCB and the water-soluble chitosan at the same time.3.To inject from human hematopoietic stem in Sub-lethally irradiated mice is a probable way of medicine that immunity that used for the analysis device strengthen the function.
