Objective:To explore the quality and number of hematopoietic stem/progenitor cells as well as immunophenotypic features of lymphocytes in human umbilical cord blood.Methods: analysis of the capacity of colony forming unit was performed by means of methylcellulose culture method and the immunophenotype of hematopoietic stem/progenitor cells and lymphocyte in human umbilical cord blood was analysed by flow cytometry using a large panel of McAb in double labelling in 34 human umbilical cord blood samples.Results:In the 34 human umbilical cord blood samples,the recovery rate of nucleated cells was 79.2%?13.7% and the average total nucleated cell count was (12.2?.3) X 108.In methylcellulose medium,the average total colony yield of CFU-E and CFU-GM were (2.05 ?1.22) X 106 and (3.14 ?1.43) X 106, respectively.The average number of CD34~ce1ls was (1.98 ?1.30) X i07.In human umbilical cord blood,the total T lymphocyte count and CD8~T lymphocyte count were less than that抯 of adult peripheral blood (P<0.Ol and P<0.001, respectively.) ,the CD4-CD8 ratio was more than that in adult peripheral blood (P<0.002) and the large majority of human umbilical cord blood T lymphocytes co-expressed CD45RA naive antigen, the proportion of CD4~CD45RA~ cells was 90.97% ?6.96% and that of CD8~CD45R.A~ cells was 98.93% ?0.36%.There were significant differences in percentages between in human umbilical cord blood and in adult peripheral blood I lymphocytes(P<0.001).Conclusion:The nucleated cells in single human umbilical cord blood can meet the need of transplantation for one adult,the total I lymphocyte count and CD8~T lymphocyte count were less in human umbilical cord blood than that抯 of adult peripheral blood,morever,the large majority of human umbilical cord blood T lymphocytes were naive T cells,the immunologic function of lymphocytes in human umbilical cord blood was immature,their immunoreactive potential to homogeneity is weaker,so the incidence and severity of Graft-versus-Host Disease (GVHD) may be low 4 after umbilical cord blood transplantation.
