ObjectiveTo investigate the relationship between expression of TNF- α,NF-κB in drowning rat brain tissues and brain injury after stress, which induced by being drowned in fresh water. The research will provide theoretic evidence for applying blocker of cytokine to pretreat brain injury and lower the happening of nerve sequela in the clinical treatment of drowning patients.Method1. 70 Wistar rats were divided into three groups randomly: group Ⅰ (10 healthy rats);group Ⅱ(10 rats were drowned in fresh water untill they all died);group Ⅲ(50 rats, each 10 rats were killed respectively at hour 3,6,12,24,40 after being drowned in fresh water);the related index of all the animals were measured according to requisition of experiment, all samples of brain tissues were obtained and prepared for pathologic detection.2. The fixed brain tissues were sectioned and evaluated by normal hematoxyline-eosin staining.3. To measure the expressions of tumor necrosis factor-a and nuclear factor- K B in brain tissues with the method of immunohistochemistryResult1. Pathologic changes of brain tissues :group I :the brain structure was normal without any evidences of edema, hemorrhage and necrosis;group II :the brain essentiality were loose ,the diastem around small blood vessels became enlarged, parts of the nerve cell became larger accompanied with degeneration. groupHI:degree of brain injury increased gradually .the worst injury appeared at 40 hour time point after being drowned, iquefaction necrosis were found in the brain essentiality.2. It was significant that brain modulus in group I were lower compare with those of group II (p<0.05),in which brain modulus were greater than that of hour 3 and hour 12(p<0.05);There were no statistic significance between the brain modulus at hour 24 ,40 and group II (p >0.05) .3. Vague expressions of TNF- a were observed in group I and group II;in group III, the expression of TNF- a gradually rose from hour 3 and reach its peak at hour 12, which were significant compared with that of group I (p<0.05).4. The expressions of NF- K B in group II were higher than that of group I . During the course of dynamic detection which began from hour 3,the expressions of NF-KB gradually increased and reached the highest value at hour 6 . Compared with group I ,the expressions of NF-KB at hour 6 and hour 12 were significant(p<0.05).5. Positive correlation were found between the expression of NF-KB and TNF- a in the injuried brain tissues after being drowned.Conclusion1. The brain of survivals after being drowned in the fresh water may be injuried badly , high expressions of TNF- a and NF- K B were found during the injury.2. The cascade amplification effects of NF- K B and TNF- a play an important role in brain injury after being drowned.3. The results suggest that proper measures which antagonize the excessive production of NF- K B and TNF- a will play positive role in preventing brain injury in clinic.
