ObjectiveOn the basis of comprehensive therapy with acupuncture for apoplexy at the present time,combining modern pharmacological research to prepare collagen tetramethylpyrazine sustained-elease dose,to research the effect of embedding medicine in Dazhui and Neiguan acupoints on the infarct volume and its regulating effect on tumor necrosis factor-αand nuclear factor kappa B.MethodsTotally 160 healthy SD rats were selected and divided randomly into 6 groups,The groups included normal group,sham group,model-24h group, model 72h group,model 120h group,abdominal cavity injection 24h group, abdominal cavity injection 72h group,abdominal cavity injection 120h group,electroacupuncture 24h group,electroacupuncture 72h group, electroacupuncture 120h group,embedding medicine in acupoints 24h group,embedding medicine in acupoints 72h group,embedding medicine in acupoints 120h group,with 10 in each group.The model of ischemia reperfusion of cerebral middle artery was established by the modified thread occlusion method,then embedding medicine in Dazhui and Neiguan acupoints with collagen tetramethylpyrazine sustained-release dose.4 rats of each group were decollated directly to investigate the expression of TNF-αin hippocampus by radioimmunoassay and the expression of NF-κB- p65 protein in hippocampus were assayed by immunohistochemistry;and other 6 rats were reperfused with 4% paraformaldehyde after being anaesthetized by 10%chloral hydrate,and then taken out the brain rapidly.The brain tissue 2mm away from the chiasma was cut frontally,the middle part left,immersed in hard paraffin.Used immunhistochemistry SABC method to investigate the expression of NF-κB,and the effect of all the above index of electro-acupuncture were observed.The entire datum was analyzed by statistics.Results1.MACO and reperfusion injury can damage the rats activity distinctly.The nerve symptoms of abdominal cavity injection group,electro-acupuncture group and embedding medicine in acupoints group were ameliorated obviously;there was a significant difference between model group and each therapy group,and the embinding medicine in acupoints group can improve rats nerve sympotom remarkably furthermore(compared with abdominal cavity injection group,P<0.01; compared with electro-acupuncture group,P<0.05).2.The expression of radioimmunoassay,after ischemia 30mins and reperfusion 24h,the content of TNF-αin hippocampus of model group rats reached peak value and then dropped gradually,which was significantly higher than those of normal and sham operation group rats.Compared with model group,the content of TNF-αin hippocampus of each treatment group was obviously lower(P<0.01)and then decreased with the lapse of time.The effect of embedding implantation of collagen and tetramethylpyrazine in points was excelled the group of injecting abdomen distinctly(P<0.01).3.In comparison with normal group,NF-κB- p65 expression positive cells of hippocampal CA1 area in 24h of model group,and every 72h and 120h subgroup of model and embedding medicine in points were significantly more(P<0.05,).Compared with 24h,72h,120h subgroup of model group,NF-κB- p65 expression positive cells in the corresponding subgroups of embedding medicine in points group were significantly fewer(P<0.01).Also along with pours into the time to change again,the expression of 72h was to reach height,NF-κB is distinct transported to the nuclear.While the grey values of NF-κB-p65 in the 3 subgroups of embedding medicine in points group were markedly lower than those in the corresponding 3 subgroup of model group.ConclusionThe expression of TNF-αincreased and the expression-positive cells of hippocampal CA1 area of NF-κB- p65 protein increased in ischemical brain tissue after focal cerebral ischemia reperfusion in rats,which computated the inflammation reaction,and incurred the neuronal cells damnification by inflammation reaction.Embedding Medicine in Points can reduce the expression of TNF-αand NF-κB-p65 protein in hippocampus and prevent it from being transported to the nucleus of the neurons in hippocampal CA1 area,which may contribute to the protective effect of on ischemic neurons.
