Objective: To investigate correlations between expression of MMP-9,EGFR, MVD and clinicalbiological characteristics in non-small cell lung cancer.Methods: High sensitive S-P immunohistochemical method was used to detect the expressions of MMP-9, EGFR and MVD in 65 cases of NSCLC and 18 cases of normal lung tissues. To study the relationships between their expressions and histological types of NSCLC,Tumor size,TNM stages, differentiations, lymph node metastasis and their correlations each other.Results: (1) The positive rates of MMP-9 expression was 63.1% in NSCLC, which was dramatically higher than that in a normal tissues(16.7%) ( P<0.05) (2) Firstly, the positive rate of MMP-9 expression was 61.1% in squamous cell carcinomas,65.2% in adenocarcinomas and 66.6% in big cell lung carcinomas respectively. There was no correlation with the histological types of NSCLC (P> 0.05) . Secondly, the positive rate of MMP-9 expression was 58.9% in tumor size=4cm and 69.2% in tumor size>4cm, There was no correlation with tumor size of NSCLC( P>0.05 ) .Thirdly, the positive rate of MMP-9 expression was 53.5% in stage I +II and 81.8% in stage III+IV respectively, which was significant relationship with TNM stages (P<0.05). Fourth, the positive rate of MMP-9 expression was 47.4% in well differentiations and 85.1% in poor differentiations. There was closely correlation with differentiations (P<0.05) Lastly, correlated with lymph node metastasis, the positive rate of MMP-9 expression was 48.5% in tissues of no lymph node metastasis and was78.1% in tissues of lymph node metastasis . The expressionof MMP-9 was highly correlation with lymph node metastasis (P<0.05) . Therefore, the MMP-9 expression in NSCLC was significantly correlated to TNM stage, differentiations and lymph node metastasis. It was no relationship with the histological types and tumor size of NSCLC. (3) The positive rates of EGFR expression was 53. 8%in NSCLC, which was dramatically higher than that in normal tissues(5.6%) ( P<0.05) (4) Firstly, the positive rate of EGFR expression was 52.8% in squamous cell carcinomas,56.5% in adenocarcinomas and 50.0% in big cell lung carcinomas respectively. There was no correlation with the histological types of NSCLC (P>0.05) . Secondly, the positive rate of EGFR expression was 46.1% in tumor size=4cm and 69.2% in tumor size>4cm, There was no correlation with tumor size of NSCLCC P > 0.05 ) .Thirdly, the positive rate of EGFR expression was 44.2% in stage I + II and 72.7% in stage HI + IV respectively, which was significant relationship with TNM stages (P<0.05). Fourth, the positive rate of EGFR expression was 42.1% in well differentiations and 70.4% in poor differentiations. There was closely correlation with differentiations (P<0.05) Lastly, correlated with lymph node metastasis, the positive rate of EGFR expression was 39.4% in tissues of no lymph node metastasis and was 68.6% in tissues of lymph node metastasis . The expression of EGFR was highly correlation with lymph node metastasis (P<0.05) . Therefore, the EGFR expression in NSCLC was significantly correlated to TNM stage, differentiations and lymph node metastasis. It was no relationship with the histological types and tumor size of NSCLC. (5) In NSCLC MVD was 41. 30 ± 16. 77, which was dramatically higher than that in normal tissues(10. 44 + 5. 64) ( P<0.05) (4) Firstly, MVD was 40. 27 + 16.16 in squamous cell carcinomas, 42. 26 + 18.11 in adenocarcinomas and 44.33+17.98 in big cell lung carcinomas respectively. There was no correlation with the histological types of NSCLC (P> 0.05) . Secondly, MVD was 39. 70 + 15. 65 in tumor size=4cm and 45. 64+16. 32 in tumor size>4cm, There was no correlation with tumor size of NSCLC (P > 0.05) .Thirdly,MVD was 35. 93 + 14. 57 in stage I + II and 51. 95 +16. 09 in stage III + IV , respectively, which was significant relationship with TNM stages(P<0.05) .Fourth,MVD was 33. 73±14. 05 in well differentiations and 52. 70± 14. 51 in poor differentiations. There was closely correlation with differentiations(P<0.05) Lastly, correlated with lymph node metastasis, MVD was 34. 51 ± 14. 63 in tissues of no lymph node metastasis and was48. 40 ±16.12in tissues of lymph node metastasis .MVD was highly correlation with lymph node metastasis (P<0.05). Therefore,MVD in NSCLC was significantly correlated to TNM stage, differentiations and lymph node metastasis. It was no relationship with the histological types and tumor size of NSCLC.Conclusion: MMP-9,EGFR play important roles in non-small cell lung cancer procession,especially in tumor metastasis.MMP-9 and EGFR have intimate relationship with MVD.
