Object To investigate the change of protein expression of MMP-2and E-cadherin in benign and malignant disease of esophagus and toanalysis the relationship between two molecules with tumorigenesisand metastasis of esophageal squamous cell carcinoma by correlatingthe results with clinicopathologic variables.Methods Expression of MMP-2 and E-cadherin in fifty cases of normalesophageal mucosa, and forty-five cases of esophageal squamous cellcarcinoma were determined by S-P immunohistochemical method.Results Protein expression of MMP-2 was detected in a significantproportion of esophageal squamous cell carcinoma(76.6%) tissuesthan normal tissues(13.3%)(P<0.01). The high level expression ofMMP-2 protein was strongly correlated with clinical stage, lymphnode metastasis, depth of infiltration in esophageal carcinoma.The expression of E-cadherin protein in esophageal carcinoma(17.8%)was significantly lower than normal tissue(86.7%) (P<0.01). Wefound a significant correlation between the expression ofE-cadherin and clinical stage, lymph node metastasis, depth ofinfiltration, and tumor diferentiation. The negative relationshipbetween the expression of MMP-2 and E-cadherin could be found withstatistical significance. Conclusion The expression of MMP-2 andE-cadherin contributed to tumorgenesis in the early stage ofesophageal squamouscell carcinoma.The expression of MMP-2 andE-cadherin have close correlation with metastasis,Which may beconsidered as two potential indicators to judge the metastasis andprognosisof esophageal cancer .Because of negative relationship ofMMP-2 and E-cadherin in pathological course of esophageal cancer,combined measurement of MMP-2 and E-cadherin maybe have moreveracity than singIe Index measurement to forecast the malignantbiological character.
