Aging is an inevitable biological change process of Molecule,Cell,Organ,Physiology and Pathology.This is a synthetic,sophisticated and motive process even the whole body functiondecreasing.The aim of the study ageing is to seek for the effectiveantiageing methods, decrease the lower speeds of organ and functionas ageing and uphold the life quality to postphone arrival of ageing.The free radical theories of aging are thought the free radicalinclude the production of superoxide,hydrogen peroxide and thehydroxyl radical. They can induced molecular and cellulardamage.The antioxidative defense system that protects against theoxidative damage caused by reactive oxygen species is suppressedwith age. Therefore,the aging process is thought to lead to animbalance between oxidative damage and the antioxidative defensesystem.Many Chinese traditional medicines contains some naturalantioxidants and their less side effects and toxicity than syntheticantioxidants.Aralia elata is a tonic Chinese medicinal and containsbiological active materials,such as soponins,fats votatileoil,etc.Pharmacology experients proved that total soponions canresist myocardial anoxemia,protect liver,decline blood fat,tolerateoxygen lack,anticancer and antiflammation .The Objective of this experimemtal studies is to set D-galactoseinduced aging model, probe effects of anging mice heart and brainbetween groups to certify the antiaging effect of the aralia elata anddiscuss the antioxidant effect and the biological mechanisms ofaging.The experimemt uses male and female Kunming miceassigned to five groups.That is normal group,model group,smallgroup(7mg/kg.d) , medium group(14mg/kg.d) and large group(28mg/kg.d) at random.The normal group are given same dose NaCl(0.9%),The othergroups are given injecting D-galactose under back skin in order toset ageing model and are offerd different dose Aralia elata throughstomach.The organs of the mice are taken after 60 days.Usingbiochemics methes measure the content of superoxide dismutase(SOD)and malondialdehyde (MDA)of the mice in their cerebralcortex and heart. Using normal histological HE stain and electronmicroscope technology observe the common morphology andultrastructure changes of the mice in their cerebral cortex and heart.The biochemics experimemtal effects show that model groupcan increase MDA and reduce their SOD of the mice heart andcerebral cortex comparing the normal group.It has obvious statisticaldifference(P<0.05).Comparison with D-galactose group showed that,the content ofMDA of administration of aralia elata in the mice heart and cerebralcortex lowered, the activity of SOD increased . It has obviousstatistical difference(P < 0.05) 。 The morphology experimemtaleffects show, the number of the heart muscle in the model group areless than that of the normal group in the optical microscope;Theneure of the cerebral cortex are in constriction than the normal.andwe can see some death cell.In the electron microscope, changes incardiac muscle including: part of cell nucleus occurfracture;sarcomere construction disperse;intercalated disc aredisorganized. The nucleus of neure have irregular outlines,;nuclearmatrix are hollow;around the nucleus rough endoplasmic reticulum,free ribosome ,Golgi complex and mitochondria decrease andlipofuscin granules increase. The number of the synapse in theneuropil have decreased. There are number less neurofilament andmitochondrion in the process.we can see the phenomenon of theneuronphagla.Comparing with the model group,ther are moe heartcells and regular in the all Aralia elata groups in the opticalmicroscope;The neurone increase , there are round nuclear,nucleolus are obvious,constriction neurone decrease and we can notsee the the neuronphagla. Especially ,this phenomenon is moreobvious in the medium and large groups.Conclusion from the experimemt:D-galactose could induce todevelop changes analogue to aging in the mice brain and heart.Araliaelata can increase SOD and reduced their MDA in the heart andcerebral cortex of the mice. Th brain and heart of decrepit micecaused by D-glactods and Aralia elata could ameliorate obviouslythe fade the common morphology and ultrastructure changes of brainand heart in the aging process. Aralia elata can increaseanti-oxidation ability of decrepit mice by D-glactods and has aneffect of anti-senility through improving free radicals. Aralia elatacould postphone antiaging process by raising the antioxidantcapacity,lowering the lipid peroxidation.
