Study On Immunomodulation And Tumor-inhibition Of Bifidobacterium Cell Wall In Mice

Objective To explore the immunologic regulation of the bifidobacterium adolescence cell wall(BCW) on function of macrophage in mice and its inhibition of BCW on human transitional cell carcinoma of bladder(BTCC) in vivo and in vitro. Methods BCW were isolated and injected into abdomen of KM mice,put the mice to death after seven days,gather the macrophage under the condition of asepsis,the level of IL-1,IL-12 and TNF-a which produced by macrophage were determined by ELISA. Cultivated the cell of human transitional cell carcinoma of bladder(T₂₄) until the cell was in the stage of logarithmic growth established the model of T₂₄ mice, then BCW was injected in abdomen of KM mice which had been established T24, after fourteen days, peel off the tumor tissue, weight,and calculated the tumor-inhibition rate;the role of tumor inhibition in vitro was determined by MTT method, gather the macrophage under the condition of asepsis,the level of IL-1,IL-12 and TNF-a which produced by macrophage were determined by ELISA. Results The value A of IL-1 in BCW injection group was (0.62±0.04), while the control group was (0.34±0.02),the level of IL-12 in BCW injection group was (823.34±185.32)pg/ml,while the control group was (498.26±105.43)pg/ml, and the level of TNF-a in BCW injection group is (1.32±4.25) ng/ml while the control group is (0.45±0.33) ng/ml.The level of IL-1,IL-12 and TNF-a with injection of BCW group was higher than those of control group(.P<0.01);After trement the size and weight in group with injection of BCW was reduced. There was significant difference between BCW injected group and control group(.P<0.01). BCW could inhibite the growth of BTCC in vitro and the tumor-inhibition become stronger with the increase of BCW concentration and become stronger with the more time(P<0.05);The value A of IL-1 in BCW injection tumor-bearing group was (0.76±0.02), while the tumor-bearing control group was (0.41±0.05),the level of IL-12 in BCW injection tumor-bearing group was (786.34±154.32)pg/ml,while the tumor-bearing control group was (520.35±89.37)pg/ml, and the level of TNF-a in BCW injection tumor-bearing group was (1.41±4.25) ng/ml while the tumor-bearing control group was (0.52±0.18) ng/ml. The level of IL-1,IL-12 and TNF-a of BCW injection tumor-bearing group were higher than those of tumor-bearing control group(p<0.01)Conclusion BCW can activate macrophage to secrete more IL-1 ,IL-12 andTNF-a;BCW can inhibite the growth of BTCC in vivo and in vitro.
