Objective To investigate the anti-tumor mechanisms and signal pathways ofBifidobacterium adolescentis in view of the signal transduction.Methods MTT assay and flow cytometer were used to detect the inhibition ofproliferation, cell cycle of colorectal tumor LoVo cells and effect of apoptosis .Theactivity of protein tyrosine kinase (PTK) in tumor were measured . The difference ofexpression of nuclear factor-κB(NF-κB)in LoVo cells before and after treatment withBifidobacterium adolescentis were detected by immnocytochemistry staining.Result After treatment with Bifidobacterium adolescentis, the growth of LoVo cellswas inhibited in time-and-dose dependent manner, the cell cycle was controlled inG0/G1. Meanwhile, large apoptosis cells appeared. And the activity of PTK anddensity of positive NF-κB cell in tumor were significantly lower than those in controlgroup.Conclusion Bifidobacterium adolescentis can inhibit the growth of colorectal tumorLoVo cells. It can introduce apoptosis of tumor cells, markedly decrease the activity ofPTK and inhibit the activation of NF-κB of colorectal in vivo.
