Objective Acute liver failure (ALF) is a kind of complex clinic syndrome which is badly endangering the health of human being and can make patients in a extremely dangerous state. Its pathological changes involve many apparatus and its mortality rate can reach 70 to 80 percents. It's very important to use Chinese drugs to protect survival hepatic cells and make hepatic regeneration. The complexity of ALF clinical presentations makes it difficult to Bianzheng Lunzhi. According to the viewpoint of TCM (Traditional Chinese Medicine), toxin and stagnation are the pathogenesis of ALF. So the preventive and therapeutic rules are detoxification and removing blood stasis. At present, the medicine to ALF is urgent need. Basing on clinic experiences of the Research Institute of Hepatic Diseases, we develop DanHuangFang which use Salviamiltiorrhiza Bge to remove blood stasis and use Rheumpalmatum L to remove the toxin. The rats were injected with thioacetamide (TAA) to induce the model of acute hepatic failure rat. By observing the change of the indexes of ALT, AST, TB, interleukin (IL-6), mitosis and c-met, the effects and probable mechanisms of Dan Huang Fang (DHF) on rats with laboratory acute hepatic failure has been explored. Methods:1.112 Wistar male rats of SPF were randomly divided into six groups: normal control group (group A), model control group (group B), PHGF group (group C), DHF group of small dose (group D), DHF group of middle dose (group E), DHF group of large dose (group F). After fasted overnight for over 14h, all rats were injected with TAA 600mgkg-1 twice at same time for two days to induce the model of acute hepatic failure rat except the normal group injected with the 0.09 salt solution hypodermically. The rats were treated with the 0.09 normal saline, DHF low, middle and high dosages, PHGF respectively every day for 3 days before TAA were injected, and 48 hours later, the blood and hepatic tissues were collected.2.Observed the mortality of the rats in 48 hours, the blood were collected to measure liver function (serum ALT, AST, TBil), interleukin(IL-6).3.Observed the pathological morphologies of hepatic tissues under opticsSupported by the Natural Science Foundation fHubei Provincial (NO. 2004ABA236)...
