Objective: To study the effect of polysaccharide of laminarin 01(L01) effect on inhibit platelet function, protect endothelia cells in vivo and the vWF secretion of endothelia cells in vivo and vitro, thus providing an experimental basis for developing a new drug for antithrombosis.Methods: (1) Effect on platelet function in vivo. Rats were divided into 4 groups randomly. Rats of model group were injected adrenalin and treated with normal saline for 1d, 2d, 3d, 4d and 5d respectively. Rats of H-L01 treatment and L-L01 treatment groups were injected adrenalin and treated with L01 50mg/kg and 10mg/kg respectively. Rats of NS control group were received normal saline instead. Blood samples were collected each from the vena cava. The platelet adhesion ratio and platelet aggregation ratio were conducted with filter methods and collodium-slides methods. (2) Effect on endothelia cells in vivo. After blood was collected from rats, aortas were excised for observation of damage degree of vessel endothelium with vWF antigen immunohistochemical studies. And the concentration of plasma vWF was measured with ELISA. (3) Effect on endothelia cells in vitro. H-L01 treatment, L-L01 treatment, model control, L01 control and normal control were administered in vitro for 24h, 48h and 72h. vWF extracellular secretion in culture supernatant were measured with ELISA and vWF intracellular contents were detected with immunocytochemistry.Results: (1) Effect on platelet function in vivo. There was a significant difference in the rats of platelet function between model rats and L01 treatment rats. The platelet adhesion ratio of L01 treatment rats was significantly less than that of model rats, especially for 4d and 5d.
