Experimental Study About The Lethal Effect Of Human Tumor Infiltrating Lymphocytes Transfected With TNF-α Gene In Gastric Carcinoma

Object: Transgen tumor infiltrating lymphocytes(transgen TIL) was constructed by transfected with tumor necrosis facor-α(TNF-α) gene into human TIL.The antitumor activity of transgene TIL was studied in vitro, and its advantage in the therapy of gastric carcinoma was probed.Methods: TIL and gastric carcinoma cell were extracted from metastatic lymphoid nodes of 15 operative gastric carcinoma patients whose diagnosis were identified by pathology and were cultivanted in vitro, Lipidosome was used to transfer TNF-αgene into TIL to construct a new type of antitumor effect cell. Count the quantity of the transgene TIL and the TIL with the counting slide and then draw the growing graph of them.Verify the expression of the purpose gene with Western blots. Mensurate the antitumor activity of transgene TIL in vitro with MTT method.Results: Among the 15 cases of transgene TIL: 2 cases have doomed during the filtration;1 case has been contaminated during the cultivation; The result of western blots showed that all of the 12 left cases showed the characteristic band of TNF protein and the gradation of band were darker than those of TIL.MTT results indicated: Among 12 cases of transgene TIL ,8 cases didn't show statistically difference in the lethal rate to gastric carcinoma as compared with TIL(p>0.05), while 4 cases had statistically difference to TIL(p<0.01). lethal rate of all the cases depent on the action time and the effect-traget rate.
